基于网络药理学与分子对接分析王百合苷干预抑郁症分子机制OA
Analysis on Molecular Mechanisms of Regalosides in Depression Intervention Based on Network Pharmacology and Molecular Docking
为探究大百合(Cardiocrinum giganteum)干预抑郁症的分子机制,以大百合鳞茎为物质基础,以王百合苷A(Regalo-side A)、王百合苷B(Regaloside B)和王百合苷F(Regaloside F)为配体,在数据库中筛选大百合活性成分靶点及与抑郁症和神经退行性疾病相关靶点,并对活性成分靶点基因集和疾病相关靶点基因集进行交集运算,获得共有靶点;构建蛋白质相互作用(PPI)网络,通过拓扑特征评价筛选核心靶点,进行GO功能注释和KEGG通路富集分析;通过分子对接验证大百合活性成分与核心靶点的结合活性.结果表明,共获得94个共有靶点.ALB、SRC、CASP3、TLR4、MAPK14和HSP90AA1均为核心靶点.大百合活性成分药效主要涉及应激反应调节和神经细胞稳态维持及血清素能突触、MAPK信号通路和细胞凋亡通路.王百合苷A、B和F与MAPK14、CASP3和HSP90AA1等靶点的蛋白质均具有良好结合活性.大百合通过多活性成分、多靶点和多通路协同作用发挥抗抑郁效应,其作用机制可能与调节神经传递、抑制炎症和阻断神经元凋亡等密切相关.
In order to investigate molecular mechanisms of Cardiocrinum giganteum in dpression intervention,taking C.giganteum bulbs as material basis and Regaloside A,Regaloside B and Regaloside F as ligands,C.gi-ganteum active compound targets and related targets with depression and neurodegenerative diseases were screened from databases.Mutual targets were obtained by intersection operation between gene set for active compound targets and gene set for disease related targets.Protein-protein interaction(PPI)network was con-structed to screen core targets by evaluation of topological characteristics.Gene Ontology(GO)function annota-tion and KEGG pathway enrichment analysis were performed.Molecular docking was employed to verify bind-ing activities among active compounds and core targets.Results showed that a total of 94 mutual targets were obtained.ALB,SRC,CASP3,TLR4,MAPK14 and HSP90AA1 were core targets.Pharmacological effects of C.giganteum active compounds primarily involved regulation of stress responses,maintenance of neuronal homeo-stasis,serotoninergic synapses,MAPK signaling pathway and apoptosis pathway.Regalosides A,B and F exhib-ited strong binding activities with proteins of MAPK14,CASP3 and HSP90AA1.C.giganteum exerted synergis-tic anti-depressant effects through multi-active compounds,multi-targets and multi-pathways,with mecha-nism might closely relating to modulating neurotransmission,suppressing inflammation and inhibiting neuronal apoptosis.
