八宝丹对5-氟尿嘧啶所致大鼠心脏损伤的保护作用OA
Cardioprotective Effect of Babao Dan against 5-Fluorouracil-Induced Myocardial Injury in Rats
目的 探讨八宝丹(Babao Dan,BBD)对5-氟尿嘧啶(5-FU)所致大鼠心脏损伤的保护作用及机制.方法 将30只雄性Wistar大鼠按照随机数字表法分为对照组、模型组和治疗组,每组10只.对照组予以生理盐水腹腔注射,模型组予以5-FU腹腔注射3周,治疗组在模型组的基础上,同时给予BBD灌胃3周.采用心脏超声检测射血分数(EF)、左室短轴缩短率(LVFS),联合分光光度比色法检测肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH),综合评估心功能变化;HE染色观察心脏病理形态变化;TUNEL染色检测心肌细胞凋亡情况;透射电镜与线粒体红色荧光探针(Mito-Tracker Deep Red FM)染色评估心肌细胞线粒体损伤情况.结果 与对照组比较,模型组EF和LVFS均降低(P<0.05),CK、CK-MB和LDH水平均升高(P<0.05);与模型组比较,治疗组EF和LVFS均上升(P<0.05),CK、CK-MB和LDH水平均降低(P<0.05).HE染色结果显示:与对照组比较,模型组大鼠心肌细胞明显损伤,心脏组织间隙增大、细胞排列紊乱等病理情况;与模型组比较,治疗组大鼠心脏组织的间隙较小,细胞排列紊乱损伤程度有所缓解.TUNEL染色结果显示:与对照组比较,模型组大鼠心肌细胞凋亡率提高(P<0.05);与模型组比较,治疗组心肌细胞凋亡率降低(P<0.05).透射电镜与Mito-Tracker Deep FM染色结果显示:与对照组比较,模型组大鼠心肌组织线粒体明显减少,荧光强度变弱(P<0.05);与模型组比较,治疗组大鼠心肌组织线粒体数量上升(P<0.05),但荧光强度变化差异无统计学意义(P>0.05).结论 5-氟尿嘧啶对大鼠心脏具有明显的毒性作用,包括心功能下降、心肌受损、心肌线粒体减少;八宝丹能够减轻5-氟尿嘧啶引起的心脏毒性作用,对心脏具有保护作用.
Objective:This study aims to investigate the cardioprotective effect of Babao Dan(BBD)against 5-fluorouracil(5-FU)-induced cardiac injury in rats,and to explore the underlying mechanism.Methods:Thirty male Wistar rats were randomly divided into three groups:the control group,the model group,and the treatment group,with 10 rats in each group.Rats in the control group received an intraperitoneal(i.p.)injection of normal saline;rats in the model group received an i.p.injection of 5-FU for 3 consecutive weeks;rats in the treatment group received 5-FU injection combined with concurrent intragastric administration of BBD for 3 consecutive weeks.Cardiac function was evaluated via echocardiography to assess ejection fraction(EF)and left ventricular fractional shortening(LVFS),in combination with spectrophotometric colorimetric assays to detect levels of creatine kinase(CK),creatine kinase-MB isoenzyme(CK-MB),and lactate dehydrogenase(LDH).Histopathological changes in cardiac tissue were assessed via hematoxylin-eosin(HE)staining.Myocardial apoptosis was detected via terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)staining.Car-diomyocyte mitochondrial damage was evaluated via transmission electron microscopy(TEM)and MitoTracker Deep Red FM staining.Results:Compared with the control group,the model group exhibited a significant decrease in EF and LVFS(P<0.05),along with re-duced levels of CK and CK-MB(P<0.05).Compared with the model group,the treatment group showed significantly increased EF and LVFS(P<0.05)and decreased LDH levels(P<0.05).HE staining revealed severe myocardial injury in the model group,characterized by widened interstitial spaces and disorganized cardiomyocyte arrangement,while these pathological changes were alleviated in the treat-ment group.TUNEL staining demonstrated that the number of apoptotic cardiomyocytes was significantly higher in the model group than in the control group(P<0.05),and this elevation was significantly attenuated in the treatment group(P<0.05).TEM combined with MitoTracker Deep Red staining showed that mitochondrial count and fluorescence intensity were significantly lower in the model group than in the control group(P<0.05);compared with the model group,the treatment group had an increased mitochondrial count,but the change in fluorescence intensity did not reach statistical significance(P>0.05).Conclusion:5-Fluorouracil induces significant cardio-toxicity in rats,which is characterized by impaired cardiac function,myocardial injury,and reduced myocardial mitochondrial content.Babao Dan can attenuate 5-FU-induced cardiotoxicity and exert cardioprotective effects.
