减蛋综合征病毒纤维蛋白和六邻体蛋白可溶性表达条件的优化OA
Optimisation of Conditions for Soluble Expression of Egg Drop Syndrome Virus Fiber and Hexon Proteins
为实现减蛋综合征病毒(Egg drop syndrome virus,EDSV)AV127株纤维蛋白(Fiber)、五邻体蛋白(Penton)和六邻体蛋白(Hexon)在原核系统中的可溶性表达,用基因克隆法将病毒Fiber、Penton和Heron基因分别克隆至原核表达载体pET-32a(+)的多克隆位点上,构建了重组质粒pET-32a(+)-Fiber、pET-32a(+)-Hexon 和 pET-32a(+)-Penton,并转化至表达菌株 Rosetta(DE3)中进行诱导表达,对 Fiber和Hexon蛋白进行可溶性表达条件优化.SDS-PAGE结果显示,Penton蛋白可溶性表达量最高,占全菌蛋白的95.30%,Hexon蛋白通过优化IPTG的浓度,可溶性表达量从63.29%提高至83.73%.Fiber蛋白通过优化表达载体、诱导温度、时间和表达菌株等,最终Fiber的可溶性表达量从28.54%提高至75.04%.本研究成功建立了 EDSV主要结构蛋白的高效表达体系,为蛋白纯化和功能研究奠定了基础.
To achieve soluble prokaryotic expression of Fiber,Penton and Hexon proteins of Egg Drop Syn-drome Virus(EDSV)strain AV127,the genes encoding these proteins were cloned into the multiple clo-ning site of the prokaryotic expression vector pET-32a(+),generating recombinant plasmids pET-32a(+)-Fiber,pET-32a(+)-Hexon,and pET-32a(+)-Penton.Then,these three recombinant plasmids were transformed into Rosetta(DE3)for induction expression,and the soluble expression conditions of Fiber and Hexon proteins were further optimized to achieve the maximum soluble expression level.The SDS-PAGE i-dentification results showed that Penton protein had the highest soluble expression level,accounting for 95.30%of the total bacterial protein.Hexon protein increased its soluble expression level from 63.29%to 83.73%by optimizing the concentration of IPTG.For Fiber protein,by optimizing the expression vector,in-duction temperature,time,and expression strain,the soluble expression level was increased from 28.54%to 75.04%,achieving the maximum soluble expression of Fiber,Penton,and Hexon proteins.This study suc-cessfully established an efficient expression system for the major structural protein of EDSV,laid a solid foundation for subsequent protein purification and functional studies,and also had important reference val-ue for the prokaryotic expression of other viral structural proteins.
