Transcriptomic Analysis for Screening Immune-Related Genes in PEDV-Infected IPEC-J2 Cells

通过RNA-seq技术分析猪流行性腹泻病毒(PEDV)LJX株感染IPEC-J2细胞后的转录组变化,筛选免疫相关基因和通路,为了解宿主免疫相关因子在PEDV感染过程中的作用提供参考.通过Real-Time PCR和间接免疫荧光技术确定PEDV LJX株在猪肠上皮细胞-J2(IPEC-J2)细胞中的复制峰值期为24 h,选择PEDV感染该时间段的细胞样本进行转录组测序,筛选差异表达基因并进行GO功能富集分析和KEGG通路富集.结果显示共筛选出135个差异显著的基因,有77个基因表达上调,58个基因表达下调.通过GO功能富集分析和KEGG通路分析筛选到4个免疫相关基因即TLR2、FOS、CLEC4D 和IL23A.其中TLR2、FOS显著富集于Toll样受体信号通路,CLEC4D 和IL23A显著富集于C型凝集素受体信号通路.进一步选取免疫相关基因TLR2、FOS、CLEC4D、IL23A等进行Real-Time PCR验证,其表达趋势与转录组测序结果一致.本研究筛选到4个免疫相关基因TLR2、FOS、CLEC4D、IL23A可能在防御PEDV感染中发挥重要作用,为探究宿主抵抗PEDV感染的免疫反应机制提供参考.

The aim of this study is to investigate the changes in the genomic transcription levels of pig epi-demic diarrhea virus(PEDV)LJX strain in vitro,which will provide a reference basis for understanding the role of host immune-related factors in PEDV effection.In this study,we analyzed the transcriptomic changes in IPEC-J2 cells infected with the LJX strain of PEDV and identified immune-related genes and pathways.The optimal infection time of PEDV in IPEC-J2 cells was determined to be 24 hours using Real-Time PCR and indirect immunofluorescence techniques.Subsequently,cell samples infected for 24 hours were selected for transcriptome sequencing to identify differentially expressed genes(DEGs).The results showed that a total of 135 DEGs were identified after PEDV infection,including 77 up-regulated genes and 58 down-regulated genes.Four immune-related genes were identified through GO functional enrichment a-nalysis and KEGG pathway analysis.KEGG analysis found that TLR2 and FOS were significantly enriched in the Toll-like receptor signaling pathway,while CLEC4D and IL23A were significantly enriched in the C-type lectin receptor signaling pathway.The expression trends of DEGs were further validated by Real-Time PCR.This study identified four innate immunity-related genes(TLR2,FOS,CLEC4D,and IL23A)that play crucial roles in defending PEDV infection,providing insights into the mechanisms of host immune responses to PEDV intection.
