四倍体冰草属种质资源的表型综合评价与核心种质构建OA

冰草属物种不仅在全球范围内被视为重要的优良牧草,更是通过远缘杂交手段拓宽小麦基因源基础、改良综合性状的关键外源供体.为促进冰草属种质资源的高效利用,以206份不同来源的冰草属种质资源(不包含沙芦草)作为研究对象,首先利用流式细胞术(Flow cytometry)结合细胞学进行倍性鉴定;随后基于16个表型性状,通过多样性分析、相关性分析、聚类分析和隶属函数分析,对四倍体冰草属种质资源进行综合分析评价;最后从由2种遗传距离、3种取样方法、8个取样比例及8种聚类方法组合的384种不同策略中,筛选构建四倍体冰草属核心种质的最佳方案,并验证其代表性.结果表明,供试冰草属种质资源包括四倍体187份、二倍体8份、六倍体11份.四倍体冰草属种质的16个表型性状变异系数介于15.78%~38.26%,平均为24.42%;遗传多样性指数介于1.58~1.99,平均为1.83,表明四倍体冰草属种质资源具有较高的表型多样性.通过系统比较,确定"欧氏距离+多次聚类优先取样法+15%取样比例+最短距离法"作为构建四倍体冰草属核心种质的最佳策略,并由此构建了包含28份资源的核心种质.多维度的验证结果表明,核心种质能够有效代表原始种质的遗传特征和群体结构.通过隶属函数综合评价筛选出综合得分较高的5份优异种质:Z559、Z1511、LF112、Z1780、LF1782,均包含在核心种质群体内,进一步证明了核心种质的优异性.本研究构建的四倍体冰草属核心种质能较为完整地代表原始种质群体,为冰草属种质资源的保存、评价及遗传改良提供了理论依据和材料基础.

Species of the genus Agropyron Gaertn.are not only recognized as premium perennial forages worldwide but also serve as critical exogenous donors for broadening the genetic base and improving the comprehensive traits of wheat through wide hybridization.To promote the efficient utilization of Agropyron germplasm resources,this study investigated 206 accessions of diverse origins(excluding A.mongolicum).Ploidy levels were first identified using flow cytometry in combination with cytological observations.Subsequently,a comprehensive evaluation of tetraploid Agropyron germplasm was performed based on 16 phenotypic traits through diversity,correlation,cluster,and membership function analyses.Finally,the optimal strategy for constructing a tetraploid Agropyron core collection was screened from 384 combinations involving two genetic distances,three sampling methods,eight sampling proportions,and eight clustering methods,followed by a representativeness validation.The results indicated that the tested germplasm consisted of 187 tetraploids,8 diploids,and 11 hexaploids.The coefficient of variation for the 16 phenotypic traits in the tetraploid accessions ranged from 15.78%to 38.26%,with an average of 24.42%.The genetic diversity index ranged from 1.58 to 1.99,with an average of 1.83,indicating high phenotypic diversity.Through systematic comparison,the strategy of"Euclidean distance+multiple cluster priority sampling+15%sampling proportion+single distance method"was identified as the optimal strategy,leading to the construction of a core collection containing 28 accessions.Multi-dimensional validation confirmed that the core collection effectively represents the genetic characteristics and population structure of the initial collection.Five elite germplasm accessions with high comprehensive scores,namely Z559,Z1511,LF112,Z1780 and LF1782,were screened out via subordinate function comprehensive evaluation.All of them were included in the core germplasm collection,which further verified the superiority of the core germplasm.In summary,the tetraploid Agropyron core collection developed in this study provides a reliable representation of the original population,offering a theoretical basis and material foundation for the preservation,evaluation,and genetic improvement of Agropyron germplasm.
