类器官模型的构建及其在中药研究中的应用分析OA
Construction and application of organoid models in traditional Chinese medicine research
中药研究长期面临成分复杂、作用机制多靶点,以及整体性效应难以深入阐释等技术瓶颈,亟需建立更接近体内生理病理状态的新型研究模型与平台.近年来,类器官技术作为一种新兴的三维培养体系,已在药物筛选、毒性评价及个体化医疗等领域得到广泛应用,为中药研究提供了新的技术手段与研究视角.然而,类器官模型在中药研究中的应用尚处于起步阶段,相关研究案例与系统性总结仍显不足.该文系统综述了不同类型类器官模型的构建特征及其在中药研究中的应用价值,重点归纳了类器官在中药、中药复方、中药毒性和安全性评价中的应用,包括在作用机制解析、活性成分筛选、毒性及安全性评价、复方配伍规律探讨以及中药新药研发等方面的应用价值.同时,针对当前类器官技术在标准化构建、复杂微环境模拟、功能稳定性维持及与中药复杂体系适配,特别是"病证结合"模型等方面存在的挑战与局限,提出相应优化思路,并展望其与微流控技术、人工智能、3D 生物打印及多组学分析等前沿技术交叉融合在破解中药研究复杂性难题中的应用前景,以期为深入推进中药现代化研究提供系统参考和理论借鉴.
Traditional Chinese medicine(TCM)research has long faced technical bottlenecks,including the complexity of chemical composition,multi-target mechanisms of action,and the difficulty of deeply elucidating holistic effects.There is an urgent need to establish novel research models and platforms that more closely recapitulate in vivo physiological and pathological states.In recent years,organoids,as emerging three-dimensional culture systems,have been widely applied in drug screening,toxicity evaluation,and personalized medicine,offering new technical tools and perspectives for TCM research.However,the application of organoid models in TCM research is still in its early stage,and relevant case studies and systematic summaries remain insufficient.This article systematically reviews the construction characteristics of different types of organoid models and their application value in TCM research,with a focus on the applications of organoids in the studies of medicinal herbs,TCM formulas,and TCM toxicity and safety evaluation.These applications include elucidation of mechanisms of action,screening of active components,toxicity and safety assessment,exploration of formula compatibility patterns,and development of novel TCM drugs.In view of the current challenges and limitations of organoid technology in standardized construction,simulation of complex microenvironments,maintenance of functional stability,adaptation to the complexity of TCM systems,and particularly the development of disease-syndrome combination models,this paper proposes corresponding solutions.Furthermore,this paper envisions the application prospects of integrating organoids with cutting-edge technologies such as microfluidics,artificial intelligence,3D bioprinting,and multi-omics analysis to address the complexities of TCM research,aiming to provide a systematic reference and a theoretical basis for advancing the modernization of TCM.
