超声辅助低共熔溶剂绿色提取葛花黄酮:工艺优化、机制探索与抗氧化活性研究OA
Process optimization,mechanism exploration,and antioxidant activity study of green extraction of flavonoids from Pueraria lobata flower by ultrasound-assisted deep eutectic solvent
该研究建立了一种超声辅助低共熔溶剂提取(DES-UAE)技术,用于从葛花中绿色高效提取黄酮类成分.通过制备12 种低共熔溶剂(DES),筛选出最佳溶剂体系;采用单因素试验结合响应面法优化提取工艺,并评价提取物的抗氧化活性.利用傅里叶红外光谱(FTIR)和热重分析(TG)对最佳低共熔溶剂氯化胆碱-乳酸(ChCl-Lac)进行表征,结合扫描电镜(SEM)与σ 剖面分析,探讨 DES-UAE 的提取机制.同时,应用 AGREE、AGREEprep、GAPI 和 SPMS 绿色评价体系,评估该方法的绿色性和环保性.结果表明,最佳低共熔溶剂为 ChCl-Lac,最优提取条件:超声温度 70℃、超声时间 30 min、液固比 50 mL·g-1、含水量 40%、超声功率 60 W.在此条件下,葛花苷和鸢尾苷的得率分别为 70%乙醇提取的 1.22、1.26 倍,且提取物的抗氧化活性显著优于 70%乙醇组.机制研究表明,ChCl-Lac 主要通过氢键和范德华力与黄酮类成分发生相互作用,从而促进目标成分的溶出.绿色性评价结果验证了该方法的环保性与可持续性.该研究为天然药用植物活性成分的绿色高效提取提供了新的技术参考.
In this study,an ultrasound-assisted deep eutectic solvent extraction(DES-UAE)technology was established for the green and efficient extraction of flavonoids from Pueraria lobata flower.The optimal solvent system was screened by preparing 12 kinds of deep eutectic solvents(DES).The extraction process was optimized by single factor test combined with the response surface method,and the antioxidant activity of the extract was evaluated.The optimal DES,choline chloride-lactic acid(ChCl-Lac),was characterized by Fourier transform infrared spectroscopy(FTIR)and thermogravimetric analysis(TGA).The extraction mechanism of DES-UAE was discussed by scanning electron microscopy(SEM)and σ profile analysis.At the same time,green evaluation systems of AGREE,AGREEprep,GAPI,and SPMS were used to evaluate the greenness and environmental friendliness of the method.The results show that the optimal DES is ChCl-Lac.The optimal extraction conditions are ultrasonic temperature of 70℃,ultrasonic time of 30 min,liquid-solid ratio of 50 mL·g-1,water content of 40%,and ultrasonic power of 60 W.Under these conditions,the yields of kakkalide and tectoridin are 1.22 times and 1.26 times higher than those of 70%ethanol extraction,respectively,and the antioxidant activity of the extract is significantly better than that of the 70%ethanol group.Mechanism studies show that ChCl-Lac mainly interacts with flavonoids through hydrogen bonds and van der Waals forces,thereby promoting the dissolution of the target components.The green evaluation results verify the environmental friendliness and sustainability of the method.This study provides a new technical reference for the green and efficient extraction of active ingredients from natural medicinal plants.
