基于代谢组学和转录组学研究栀子果实不同发育时期黄酮类成分积累与F3H、F3'H基因表达模式分析OA
Analysis of flavonoid accumulation and expression patterns of F3H and F3'H genes during fruit development in Gardenia jasminoides based on metabolomics and transcriptomics
为揭示栀子果实不同生长时期黄酮类物质的合成机制及关键调控基因,以栀子青果和红果的果皮与果肉为材料,采用超高效液相色谱-串联质谱(UPLC-ESI-MS/MS)技术进行非靶向代谢组学分析,结合转录组数据筛选黄酮类生物合成候选基因,并通过实时荧光定量PCR(RT-qPCR)验证其表达模式.结果表明,栀子果实黄酮类化合物在果肉中积累显著,红果肉中黄酮醇类成分尤为突出;代谢组与转录组联合分析提示芹菜素(apigenin)为关键前体,黄烷酮 3-羟化酶(F3H)和类黄酮3'-羟化酶(F3'H)是调控黄酮类化合物羟基化修饰的关键限速酶;系统发育与表达分析显示GjF3'H家族成员具有明显的进化特异性和组织表达模式.该研究通过多组学联合解析了栀子果实黄酮类物质的积累规律与合成关键基因,为栀子黄酮类成分的定向培育与质量控制提供理论依据.
To elucidate the biosynthesis mechanism of flavonoids at different developmental stages of Gardenia jasminoides fruit and identify key regulatory genes,this study used the pericarp and sarcocarp of green and red fruit as the materials.Non-targeted metabolomics analysis based on ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-ESI-MS/MS)was performed,followed by integrated transcriptomic data mining to screen candidate genes involved in flavonoid biosynthesis.The expression patterns of these genes were subsequently validated by real-time quantitative PCR(RT-qPCR).The results showed that flavonoids accumulate significantly in the pulp of G.jasminoides fruits,with flavonol components being particularly prominent in the red pulp.Integrated metabolomic and transcriptomic analysis suggested that apigenin is a key precursor,while flavanone 3-hydroxylase(F3H)and flavonoid 3'-hydroxylase(F3'H)are key rate-limiting enzymes regulating hydroxylation modifications of flavonoids.Phylogenetic and expression analyses further revealed that members of the GjF3'H family exhibit distinct evolutionary specificity and tissue-specific expression patterns.This study provides a multi-omics perspective on the accumulation patterns and key biosynthetic genes of flavonoids in G.jasminoides fruit,thereby offering a theoretical basis for the targeted breeding and quality control of flavonoid components in G.jasminoides.
