褪黑素抑制内质网应激缓解对乙酰氨基酚诱导的急性肝损伤OA

目的 探讨褪黑素在对乙酰氨基酚(acetaminophen,APAP)诱导的急性肝损伤小鼠中的保护作用及其机制.方法 以APAP(350 mg∙kg-1)构建小鼠急性肝损伤模型,并给予不同剂量的褪黑素(25、50、100 mg∙kg-1)治疗;体外以衣霉素(tunicamycin,TM)构建AML-12肝细胞内质网应激(endo-plasmic reticulum stress,ERS)模型,并给予不同浓度褪黑素或4-苯基丁酸(4-PBA)治疗.通过HE染色及血清ALT/AST检测肝组织损伤,Western blot和qPCR检测ERS相关分子表达.通过Western blot、流式细胞术和TUNEL染色检测细胞凋亡水平.结果 APAP组小鼠出现明显肝损伤,血清ALT/AST明显升高,且 ERS相关标志分子(如 PERK、IRE1α和ATF6等)的蛋白和基因表达水平明显上调.褪黑素剂量依赖性地改善肝细胞损伤并降低血清ALT/AST,且100 mg∙kg-1组效果最为明显.此外,褪黑素明显抑制APAP诱导的ERS标志分子及氧化应激水平(MDA下降、GSH上升),明显减少肝细胞凋亡.使用4-PBA抑制ERS同样降低了APAP诱导的细胞凋亡.结论 褪黑素通过抑制ERS减少APAP诱导的肝细胞凋亡,改善急性肝损伤.

Aim To investigate the protective effects of melatonin on acetaminophen(APAP)-induced acute liver injury and the underlying mechanisms.Methods An acute liver injury model was estab-lished in mice using APAP(350 mg·kg-1),followed by treatment with different doses of melatonin(25,50,100 mg·kg-1).In vitro,an endoplasmic reticulum stress(ERS)-induced model was induced in AML-12 hepatocytes using thiamphenicol(TM),treated with different concentrations of melatonin or 4-phenylbutyr-ate(4-PBA).Liver injury was assessed via HE stain-ing and serum ALT/AST levels.ERS-related mol-ecules were detected by Western blot and qPCR.Apoptosis was evaluated using Western blot,flow cy-tometry,and TUNEL staining.Results APAP-treated mice exhibited marked liver injury with signifi-cantly elevated serum ALT/AST levels and upregula-tion of ERS-related marker molecules(PERK,IRE1α,ATF6,etc.)at both protein and gene expres-sion levels.Melatonin dose-dependently ameliorated hepatocyte injury and reduced serum ALT/AST levels,with the 100 mg·kg-1 group showing the most pro-nounced effect.Additionally,melatonin significantly inhibited APAP-induced ERS markers and oxidative stress levels(decreased MDA levels and increased GSH levels),and reduced hepatocyte apoptosis.Nota-bly,4-PBA-mediated inhibition of endoplasmic reticu-lum stress also decreased APAP-induced apoptosis.Conclusion Melatonin alleviates APAP-induced acute liver injury by inhibiting endoplasmic reticulum stress and reducing hepatocyte apoptosis.
