Research progress of FGF21 in treatment of metabolic dysfunction-associated fatty liver disease
代谢功能障碍相关脂肪性肝病(metabolic dysfunction-associated steatotic liver disease,MASLD)是全球高发的慢性肝脏疾病,可引发脂肪性肝炎、肝纤维化,乃至肝细胞癌等并发症,严重威胁人类健康.由于发病机制复杂且缺乏特效治疗药物,MASLD的临床治疗面临巨大挑战.成纤维细胞生长因子21(fibroblast growth factor 21,FGF21)作为一种代谢调控激素,在调控糖脂代谢、减轻肝脏炎症与纤维化、改善胰岛素抵抗等方面发挥重要作用,近年来成为MASLD治疗领域的研究热点.该文系统综述了FGF21的生物学特性、在MASLD治疗中的作用机制、联合治疗策略及研究展望等内容,为FGF21在MASLD后续临床应用提供理论参考.
Metabolic dysfunction-associated steatotic liver dis-ease(MASLD)has become a prevalent chronic liver disease worldwide.Its progression can lead to severe complications such as non-alcoholic steatohepatitis,liver fibrosis,and even hepatocellular carcinoma,posing a serious threat to human health.Due to the complex pathogenesis and lack of specific therapeutic drugs,the clinical treatment of MASLD faces great challenges.Fibroblast growth factor 21(FGF21),as an impor-tant metabolic regulatory hormone,plays a key role in regulat-ing glucose and lipid metabolism,alleviating liver inflammation and fibrosis,and improving insulin resistance.In recent years,it has become a research focus in the field of MASLD treatment.This article systematically reviews the research progress of FGF21 in the treatment of MASLD from the aspects of FGF21's biological characteristics,mechanism of action in MASLD treat-ment,combined treatment strategies,and research prospects,so as to provide a theoretical reference for subsequent clinical application.
