目前,睡眠障碍(SD)已经成为影响全球公共卫生健康的重要问题,其发病率逐年上升.SD 发病机制复杂且尚未完全明确,涉及氧化应激、炎症反应、神经递质失衡和神经内分泌失调等多个方面.Sirt1/PGC-1α 信号通路在细胞代谢、氧化应激及线粒体功能调控中发挥关键作用,与 SD 发病机制密切相关.Sirt1作为Ⅲ类组蛋白去乙酰化酶,可通过去乙酰化修饰激活抗氧化酶、抑制炎症相关转录因子活性;PGC-1α 作为转录共激活因子,能调控线粒体生物合成、增强线粒体抗氧化能力.二者相互作用,共同调节氧化应激、炎症反应、线粒体功能以及神经递质功能,改善 SD 相关病理表现.尽管目前对 Sirt1/PGC-1α 信号通路的研究已经取得了一定进展,但仍需深入探究其在不同脑区和细胞类型中的具体作用机制.因此,需进一步优化基于该通路的药物研发,为 SD 的精准治疗提供更有力的依据.

Sleep disorders have emerged as a significant global public health issue,with their prevalence increasing annually.The underlying mechanisms remain complex and incompletely understood,involving multiple pathways such as oxidative stress,inflammatory responses,neurotransmitter imbalances,and neuroendocrine dysregulation.Sirt1/PGC-1α signaling plays a pivotal role in cellular metabolism,oxidative stress regulation,and mitochondrial function,and is implicated in sleep disorder pathogenesis.As a class Ⅲ histone deacetylase,Sirt1 activates antioxidant enzymes and suppresses the activity of inflammation-related transcription factors through deacetylation;acting as a transcriptional coactivator,PGC-1α regulates mitochondrial biogenesis and enhances mitochondrial antioxidant capacity.Their interaction jointly regulates oxidative stress,inflammatory responses,mitochondrial function,and neurotransmitter activity,improving pathological sleep disorder manifestations.Although research on the Sirt1/PGC-1α pathway has made progress,the specific mechanisms in different brain regions and cell types involved require further investigation.Optimizing drug development targeting this pathway is essential to provide stronger evidence for the precision treatment of sleep disorders.
