目的 获取树鼩卵磷脂胆固醇酰基转移酶(tsLCAT)基因的全长编码序列并进行分子特征分析.方法 从树鼩肝脏组织中提取总RNA,采用cDNA 末端快速扩增技术(RACE)克隆tsLCAT 全长cDNA;利用 MEGA 软件构建系统发育树分析进化关系,通过 ExPASy-ProtParam、SignaIP 及 MusiteDeep 等工具预测分子特征与生物学功能,使用 AlphaFold3 进行蛋白质结构预测及比对;采用 RT-qPCR 和 Western blot 检测 tsLCAT在肝脏、胰腺、肾脏等 11 种组织中的 mRNA 及蛋白表达水平.结果 获得 tsLCAT X1(全长 1 384 bp,编码440 个氨基酸)和 tsLCAT X2(全长1 400 bp,编码438 个氨基酸)两个转录变体,其中tsLCAT X1 为主要转录本.系统发育分析显示,tsLCAT 的进化关系比小鼠、大鼠等啮齿类动物更接近灵长类,tsLCAT 与人类和黑猩猩LCAT 的氨基酸序列一致性最高.组织表达谱显示,tsLCAT mRNA 在肝脏中表达量最高,蛋白质富集在肝脏和胰腺中.结论 本研究成功解析 tsLCAT 全长编码序列,系统分析其分子特征、蛋白结构及生物学功能,为树鼩作为 LCAT 相关疾病研究模型提供关键分子依据.

Objective To obtain the full-length coding sequence of the Chinese tree shrew lecithin cholesterol acyltransferase(tsLCAT)gene and carry out its molecular characterization.Methods Total RNA was extracted from Chinese tree shrew liver tissue,and the full-length cDNA of tsLCAT was cloned by rapid Amplification of cDNA ends technology.A phylogenetic tree was constructed for evolutionary relationship analysis using MEGA software.The molecular characteristics and biological functions were predicted using ExPASy-ProtParam,SignaIP,and MusiteDeep,and protein structure prediction and alignment were achieved using AlphaFold3.The mRNA and protein expression levels of tsLCAT in 11 tissues,including liver,pancreas,and kidney,were detected by reverse transcription-quantitative polymerase chain reaction and Western blot,respectively.Results Two transcript variants of tsLCAT were obtained:tsLCAT X1(full length 1 384 bp,encoding 440 amino acids)and tsLCAT X2(full length 1 400 bp,encoding 438 amino acids),with tsLCAT X1 being the major transcript.Phylogenetic analysis revealed that tsLCAT had a closer evolutionary relationship with primates than with rodents(such as mice and rats),and tsLCAT showed the highest amino acid sequence identity with human and chimpanzee LCAT.Tissue expression profiling indicated that tsLCAT mRNA expression levels were highest in the liver,and protein levels were enriched in the liver and pancreas.Conclusions The full-length coding sequence of tsLCAT was successfully obtained and characterized.Systematic analysis of its molecular characteristics,protein structure,and biological functions provides key molecular evidence for the use of Chinese tree shrews as a research model for LCAT-related diseases.
