目的:研究PLGA纳米颗粒包裹的枸杞多糖(PLGA-LBP纳米颗粒)对胶原诱导性关节炎(CIA)大鼠关节炎症的疗效及其对肠道菌群的调节作用.方法:制备PLGA-LBP纳米颗粒并评价其理化性质.构建SD大鼠CIA模型,将其随机分为正常组、模型组、LBP组、甲氨蝶呤(MTX)组、PLGA-LBP纳米颗粒组(高、中、低剂量).造模第8天开始分别用MTX、LBP、PLGA-LBP纳米颗粒治疗相应组别大鼠,35 d后,HE染色观察滑膜组织病理改变,流式细胞术分析Tfh细胞及B细胞百分比,ELISA检测血清中IgG、IL-21、IL-6 水平,16S rRNA检查大鼠肠道微生物变化.结果:与模型组相比,PLGA-LBP纳米颗粒组的滑膜病变明显减轻,可调控细菌毛螺菌属、乳杆菌属与UCG-005菌属.PLGA-LBP及LBP均能下调Tfh细胞和B细胞频数,降低血清IgG、IL-21、IL-6水平,且PLGA-LBP组效果优于LBP组.结论:PLGA-LBP纳米颗粒可缓解CIA,可能是通过调控肠道菌群减轻关节炎症.

Objective:To investigate the therapeutic effects of PLGA nanoparticle-encapsulated Lycium barbarum polysaccha-ride(PLGA-LBP nanoparticles)on joint inflammation in collagen-induced arthritis(CIA)rats and its modulatory role on gut microbiota.Methods:PLGA-LBP nanoparticles were prepared and characterized for their physicochemical properties.A CIA model was estab-lished in SD rats,which were then randomly divided into the following groups:Normal control,model,LBP,methotrexate(MTX),and PLGA-LBP nanoparticle(high-,medium-,and low-dose)groups.From day 8 post-induction,the rats in the respective groups were treated with MTX,LBP,PLGA-LBP nanoparticles.After 35 days,histopathological changes in synovial tissues were assessed by HE staining.Flow cytometric analysis of the percentages of Tfh cells and B cells.Serum levels of IgG,IL-21 and IL-6 were measured using ELISA.Additionally,alterations in the gut microbiota of rats were examined via 16S rRNA sequencing.Results:Compared with the model group,the synovial lesions in the PLGA-LBP nanoparticle group were substantially reduced.The bacteria Trichospirillum,Lactobacillus,and UCG-005 were subject to regulation.PLGA-LBP and LBP both decreased the frequency of Tfh cells and B cells,as well as serum levels of IgG,IL-21 and IL-6;this effect was more noticeable in the PLGA-LBP group.Conclusion:PLGA-LBP nanoparticles can help alleviate CIA by modifying the gut microbiota to decrease joint inflammation.
