目的:探讨玉仁雪白乳膏局部外用对黄褐斑模型小鼠皮肤"神经-内分泌-免疫"小网络功能的效应机制.方法:将40只雌性ICR小鼠随机分为空白组、模型组、10%玉仁雪白乳膏组、20%玉仁雪白乳膏组、积雪苷霜软膏组,每组8只.采用肌肉注射黄体酮联合315 nm的中波紫外线(UVB)照射的方法制备黄褐斑小鼠模型.空白组、模型组在造模区域皮肤上均匀涂抹灭菌注射用水,10%玉仁雪白乳膏组、20%玉仁雪白乳膏组、积雪苷霜软膏组,分别取1ml药膏涂抹于造模区域皮肤,每日给药1次,连续给药30 d.治疗结束后,观察记录各组小鼠造模部位皮肤色素沉着及皮肤组织病理变化情况,ELISA法检测各组小鼠皮肤组织神经系统相关因子一氧化氮(NO)、干细胞因子(Stem Cell Factor,SCF);内分泌相关因子血管内皮生长因子(VEGF)、α-黑素细胞刺激素(α-melanocyte-stimulating Hormone,α-MSH)及免疫相关因子白细胞介素-6(IL-6)、白细胞介素-10(IL-10)的表达量.结果:玉仁雪白乳膏可改善黄褐斑模型小鼠色素沉着情况,与空白组比较,模型组NO、SCF、α-MSH均明显增加(P<0.05).与模型组比较,10%玉仁雪白乳膏组、积雪苷霜软膏组、20%玉仁雪白乳膏组SCF均降低(P<0.05);10%及20%玉仁雪白乳膏组VEGF、α-MSH、IL-6均明显降低(P<0.05);10%玉仁雪白乳膏组IL-10降低(P<0.05).结论:玉仁雪白乳膏对黄褐斑模型小鼠有较好的治疗效果,其机制可能与调控黄褐斑模型小鼠局部皮肤"神经-内分泌-免疫"小网络相关生物学指标有关.

Objective To investigate the effect mechanism of Yuren snow-white cream on the function of"neuro-endocrine-immune"small network in the skin of mouse model of melasma.Methods Forty female ICR mice were randomly divided into blank group,model group,10%Yuren snow-white cream group,20%Yuren snow-white cream group and centella asiatica ointment group,with 8 mice in each group.The mouse model of melasma was prepared by intramuscular injection of progesterone combined with 315 nm UVB irradiation.The blank group and the model group were evenly coated with sterilized water for injections on the skin of the molding area,and the 10%Yuren snow-white cream group,20%Yuren snow-white cream group and centella asiatica ointment group were respectively applied with 1 ml ointment to the skin of the molding area,and were administered once a day for 30 days.After the treatment,the pigmentation of the skin and the pathological changes of skin tissue were observed and recorded.The nervous system-related factors nitric oxide(NO)and stem cell factor(SCF)and the expression of endocrine related factors vascular endothelial growth factor(VEGF),α-melanocyte-stimulating hormone(α-MSH)and immune related factors interleukin-6(IL-6)and interleukin-10(IL-10)were detected by ELISA.Results Yuren snow-white cream can improve the pigmentation of mouse model of melasma.Compared with blank group,NO,SCF and α-MSH in model group were significantly increased(P<0.05).Compared with model group,SCF in 10%Yuren snow-white cream group,centella asiatica ointment group and 20%Yuren snow-white cream group were decreased(P<0.05).VEGF,α-MSH and IL-6 in 10%and 20%Yuren snow-white cream groups were significantly decreased(P<0.05).IL-10 in 10%Yuren snow-white cream group was decreased(P<0.05).Conclusion Yuren snow-white Cream has a good therapeutic effect on mouse model of melasma,and its mechanism may be related to the regulation of the"neuroendocrine-immune"small network of the local skin of mouse model of melasma.
