大鼠颌骨和股骨骨髓来源间充质干细胞生物学特性的对比研究OA
Comparison of Biological Characteristics of Mesenchymal Stem Cells Derived from Rat Orofacial and Femur Marrow
目的:探讨大鼠颌骨和股骨骨髓来源间充质干细胞生物学特性的差异,以期为骨组织工程提供更加优质种子细胞.方法:分别从SD大鼠颌骨和股骨骨髓提取原代间充质干细胞(OMSCs和BMMSCs),流式细胞术鉴定表面标记分子,MTT、细胞划痕实验、RT-PCR、茜素红染色对比钛板刺激条件下OMSCs和BMMSCs在细胞增殖、迁移和成骨分化的差异,裸鼠异位成骨实验评估两种细胞体内骨组织形成能力.结果:OMSCs和BMMSCs均符合间充质干细胞特征,但与BMMSCs相比,OMSCs在钛板刺激下细胞增殖速率、迁移能力更强;OMSCs组矿化结节形成较多,成骨相关基因Runx2、ALP、OCN表达更高,进一步异位成骨实验显示OMSCs组新生骨量明显多于BMMSCs组,表明OMSCs成骨分化能力显著高于BMMSCs.结论:OMSCs具有更好的生物学特性,可为骨组织再生工程提供更优质的种子细胞.
Objective To explore the differences in biological characteristics between rat orofacial derived mesenchymal stem cells(OMSCs)and bone marrow mesenchymal stem cells(BMMSCs),aiming to provide higher quality seed cells for bone tissue engineering.Methods OMSCs and BMMSCs were extracted from the mandibles and femurs of SD rats,respectively.The surface marker molecules were identified by flow cytometry.Differences in cell proliferation,migration,and osteogenic differentiation between OMSCs and BMMSCs under titanium plate stimulation were compared using the MTT assay,scratch test,RT-PCR,and alizarin red staining.The ability of bone tissue formation in vivo was assessed by heterotopic ossification experiments in nude mice.Results Both OMSCs and BMMSCs met the characteristics of mesenchymal stem cells.However,compared with BMMSCs,OMSCs exhibited a stronger cell proliferation rate and migration ability under titanium plate stimulation.The OMSCs group formed more mineralized nodules,with higher expression of osteogenic-related genes Runx2,ALP,and OCN.The heterotopic ossification experiments further showed that the new bone formation in the OMSCs group was significantly more than that in the BMMSCs group,indicating that OMSCs had a significantly higher osteogenic differentiation capacity than BMMSCs.Conclusion OMSCs exhibit superior biological characteristics which can provide higher-quality seed cells for bone tissue regeneration engineering.
