目的:探讨叉头框蛋白P3(FOXP3)、高迁移率族蛋白B2(HMGB2)在结直肠癌组织中的表达,分析二者与患者临床病理特征及术后 1年复发的相关性,初步筛选结直肠癌患者术后早期复发的独立危险因素.方法:选取 2022年 1月至2025年3月接受手术治疗的70例结直肠癌患者为研究对象,采用实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测患者癌组织及癌旁组织中FOXP3、HMGB2的mRNA表达水平;通过受试者工作特征(ROC)曲线确定基因表达最佳截断值并完成分组;术后对患者开展为期1年的规范化随访,分析FOXP3、HMGB2表达与患者临床病理参数及短期预后的关系;采用单因素、多因素Cox回归模型筛选结直肠癌患者术后1年复发的危险因素.结果:经配对t检验,结直肠癌组织中FOXP3、HMGB2 mRNA表达均高于癌旁组织,P<0.05.FOXP3、HMGB2高表达与肿瘤低分化、浸润深度T3~T4期、淋巴结转移、远处转移及TNM Ⅲ~Ⅳ期密切相关,P<0.05;术后1年预后不良(复发)患者中FOXP3、HMGB2高表达占比明显高于预后良好(未复发)患者,P<0.05.单因素分析显示,淋巴结转移、TNM Ⅲ~Ⅳ期、FOXP3高表达、HMGB2高表达均与术后预后不良相关,P<0.05.多因素Cox回归分析证实,淋巴结转移、TNM Ⅲ~Ⅳ期、FOXP3高表达、HMGB2高表达是结直肠癌术后1年复发的独立危险因素,P<0.05.结论:FOXP3、HMGB2在结直肠癌组织中呈高表达状态,二者异常表达与肿瘤恶性进展及术后早期复发密切相关,有望成为评估结直肠癌恶性程度及短期预后的潜在分子标志物.

Objective To investigate the expression of forkhead box protein P3(FOXP3)and high mobility group box 2(HMGB2)in colorectal cancer(CRC)tissues,analyze their correlations with clinicopathological features and 1-year postoperative recurrence of patients,and preliminarily screen independent risk factors for early postoperative recurrence in patients with CRC.Methods A total of 70 patients with CRC who received surgical treatment from January 2022 to March 2025 were enrolled as research subjects.Real-time fluorescence quantitative reverse transcription polymerase chain reaction(qRT-PCR)was used to detect the mRNA expression levels of FOXP3 and HMGB2 in cancer tissues and paracancerous tissues of patients.The optimal cut-off value of gene expression was determined and the grouping was completed through the receiver operating characteristic(ROC)curve.The patients were followed up for 1 year after surgery,and the relation-ship between the expression of FOXP3 and HMGB2,and clinicopathological parameters and short-term prog-nosis of the patients was analyzed.Univariate and multivariate Cox regression models were adopted to analyze risk factors for 1-year postoperative recurrence of CRC.Results Paired t-test results showed that the mRNA expression levels of FOXP3 and HMGB2 in CRC tissues were significantly higher than those in paracan-cerous tissues(P<0.05).High expression levels of FOXP3 and HMGB2 were closely correlated with poorly differentiated tumors,invasive depth of T3-T4 stage,lymph node metastasis,distant metastasis and TNM stage Ⅲ-Ⅳ(P<0.05).The proportion of patients with high FOXP3 and HMGB2 expression was markedly higher in those with poor prognosis(recurrence)within 1 year after operation than in patients with favorable prognosis(no recurrence)(P<0.05).Univariate analysis revealed that lymph node metastasis,TNM stageⅢ-Ⅳ,high FOXP3 expression and high HMGB2 expression were all associated with poor postoperative prog-nosis(P<0.05).Multivariate Cox regression analysis verified that lymph node metastasis,TNM stage Ⅲ-Ⅳ,high FOXP3 expression and high HMGB2 expression were independent risk factors for 1-year postoperative recurrence of CRC(P<0.05).Conclusion FOXP3 and HMGB2 are highly expressed in CRC tissues.Their abnormal expression is closely linked to malignant progression and early postoperative recurrence of tumors,and they are expected to serve as potential molecular biomarkers for evaluating the malignant degree and short-term prognosis of CRC.
