基于NF-κB/IL-6信号通路阐释芍药汤治疗大肠湿热证溃疡性结肠炎大鼠的作用机制OA
Mechanism of Shaoyao Decoction in Treatment of Ulcerative Colitis Rats with Large Intestine Dampness-Heat Pattern Based on NF-κB/IL-6 Signaling Pathway
目的:基于NF-κB/IL-6信号通路探讨芍药汤对大肠湿热证溃疡性结肠炎(UC)大鼠的干预效果及其作用机制.方法:选用 SPF级雄性 SD大鼠 54只,先随机分出 9只为空白组,剩余 45只采用湿热环境+高脂高糖饲料+葡聚糖硫酸钠(DSS)复合法制备大肠湿热证UC模型;造模后按比例选取6只(空白组1只,造模组5只)大鼠验证造模是否成功;将剩余40只模型大鼠进一步随机分为模型组、阳性药组(美沙拉嗪组)及芍药汤低、中、高剂量组,每组8只.各组予以对应药液或等体积蒸馏水连续灌胃14 d.观察干预期间各组大鼠一般状况,记录干预后各组大鼠体质量、疾病活动指数(DAI)、结肠长度,HE染色观察结肠病理损伤并评定组织损伤指数(TDI),采用酶联免疫吸附试验(ELISA)检测血清肿瘤坏死因子-α(TNF-α)、白细胞介素-9(IL-9)、白细胞介素-10(IL-10)炎症因子水平,采用定量逆转录聚合酶链反应(RT-qPCR)与Western blot分别检测结肠组织核因子κB(NF-κB)、白细胞介素-6(IL-6)基因及蛋白相对表达量,并进行对比分析.结果:本实验成功建立大肠湿热证UC大鼠模型.与空白组相比,模型组大鼠体质量下降、DAI评分升高,结肠明显缩短,结肠黏膜破损伴大量炎症浸润、TDI评分增高,血清促炎因子TNF-α、IL-9上调,抗炎因子IL-10降低,结肠组织NF-κB、IL-6基因与蛋白表达显著升高,差异均有统计学意义,P<0.05.干预后,各给药组大鼠一般状态、肠道症状及结肠病理损伤均得到不同程度改善,DAI与TDI评分下降,结肠长度回升,血清TNF-α和IL-9水平降低、IL-10水平升高,结肠NF-κB、IL-6基因及蛋白表达被抑制(P<0.05);且芍药汤疗效呈剂量依赖性,高剂量组整体改善效果与阳性药组无明显差异(P>0.05),优于低、中剂量组.结论:芍药汤可通过抑制NF-κB/IL-6信号通路过度激活,下调促炎相关基因与蛋白表达,调控机体促炎/抗炎因子平衡,减轻结肠黏膜炎症浸润与组织损伤,从而有效治疗大肠湿热证UC,且高剂量给药干预效果更佳.
Objective To investigate the intervention effect and underlying mechanism of Shaoyao Decoction on ulcerative colitis(UC)rats with large intestine dampness-heat pattern based on NF-κB/IL-6 signaling path-way.Methods A total of 54 SPF-grade male SD rats were selected,among which 9 rats were randomly assigned to a blank group,and the remaining 45 rats were used to establish UC models with large intestine dampness-heat pattern via a combined method consisting of damp-heat environment,high-fat and high-sugar diet,and dextran sulfate sodium(DSS).After model establishment,6 rats(1 from the blank group and 5 from the model rats)were selected proportionally to verify the success of modeling.The remaining 40 model rats were randomly divided into a model group,a positive drug group(mesalazine group),and low-,medium-and high-dose groups of Shaoyao Decoction,with 8 rats in each group.Each group was intragastrically administered the corresponding liq-uid medicine or an equal volume of distilled water for 14 consecutive days.The general conditions of rats in each group were observed during intervention.After intervention,body weight,disease activity index(DAI)and colon length of rats in each group were recorded.Hematoxylin-eosin(HE)staining was applied to observe colonic patho-logical lesions and evaluate tissue damage index(TDI).Enzyme-linked immunosorbent assay(ELISA)was used to detect serum levels of inflammatory factors including tumor necrosis factor-α(TNF-α),interleukin-9(IL-9)and interleukin-10(IL-10).Reverse transcription quantitative polymerase chain reaction(RT-qPCR)and Western blot were adopted to detect the relative mRNA and protein expression of nuclear factor-κB(NF-κB)and interleukin-6(IL-6)in colonic tissues for comparative analysis.Results Rat models of UC with large intestine dampness-heat pattern were successfully established in this experiment.Compared with the blank group,rats in the model group exhibited decreased body weight,elevated DAI scores,significantly shortened colons,damaged colonic mucosa with massive inflammatory infiltration and increased TDI scores.Serum levels of pro-inflammatory factors TNF-α and IL-9 were upregulated while anti-inflammatory factor IL-10 was downregulated,and mRNA and pro-tein expression of NF-κB and IL-6 in colonic tissues were markedly increased,with statistically significant differ-ences(P<0.05).After intervention,general conditions,intestinal symptoms and colonic pathological lesions of rats in all medication groups were improved to varying degrees,accompanied by decreased DAI and TDI scores,recovered colon length,reduced serum TNF-α and IL-9 levels,elevated IL-10 levels,and suppressed mRNA and protein expression of colonic NF-κB and IL-6(P<0.05).Therapeutic effect of Shaoyao Decoction is dose-dependent.The overall improvement efficacy of the high-dose group showed no significant difference compared with the mesalazine positive drug group(P>0.05)and was superior to that of the low-and medium-dose groups.Conclusion Shaoyao Decoction can effectively treat UC with large intestine dampness-heat pattern by inhibiting excessive activation of NF-κB/IL-6 signaling pathway,downregulating expression of pro-inflammatory-related genes and proteins,regulating balance between pro-inflammatory and anti-inflammatory factors,and alleviating inflammatory infiltration and tissue injury of colonic mucosa,with better intervention efficacy at high doses.
