枣胚胎发育蛋白质分析及氨基酸代谢通路关键蛋白筛选OA

为探究不同发育时期枣胚胎中蛋白质的组成及氨基酸表达差异,采用非标记蛋白定量技术(DIA)对高度可育'酸枣'和低度可育'孔府酥脆枣'3个不同发育时期的枣胚胎进行差异蛋白GO功能注释、KEGG代谢通路注释和聚类分析.结果表明,共鉴定出10 516个蛋白,其中1 558个蛋白存在显著差异表达,1 337个具有催化活性,261个蛋白质参与氨基酸合成与代谢.GO富集分析发现,具有催化活性的差异蛋白主要富集在胚龄30 d和50 d.参与氨基酸代谢的丝氨酸乙酰转移酶(XP_015865688.1)、丝氨酸羟甲基转移酶(XP_015868935.1)和3-亚磷酸1-羧基乙烯基转移酶(XP_015888682.1)的表达量在高度可育'酸枣'胚胎发育过程中呈现持续上升趋势,在低度可育'孔府酥脆枣'胚胎中则表现为先下降再上升的趋势.特别是丝氨酸乙酰转移酶(XP_015865688.1)在'孔府酥脆枣'败育高峰期(胚龄40 d)的表达量显著下调,推测其在调控氨基酸积累中发挥关键作用.

Embryo dysplasia or abortion is a prevalent disorder in jujube(Ziziphus jujuba Mill.)em-bryos is a common phenomenon,which seriously restricts jujube cross breeding.Proteins are closely related to embryo development.Currently,systematic analyses of proteins involved in amino acid me-tabolism during embryo development in jujube varieties with different fertility levels are lacking.To explore the protein composition and amino acid expression differences in jujube embryos at different developmental stages,this study employed data independent acquisition(DIA)technology to perform Gene Ontology(GO)functional annotation of differential proteins,Kyoto Encyclopedia of Genes and Genomes(KEGG)metabolic pathway annotation,and cluster analysis of highly embryo-fertile'Sour jujube'(Ziziphus jujuba Mill.var.spinosa(Bunge)Hu ex H.F.Chow)and low embryo fertility cul-tivar'Kongfusucui jujube'at three different developmental stages.The results showed that a total of 10 516 proteins were identified,among which 1 558 proteins exhibited significant differential expres-sion.Among these differential proteins,1 337 had catalytic activity,and 261 proteins were involved in amino acid synthesis and metabolism.GO enrichment analysis revealed that the differentially ex-pressed proteins with catalytic activity were mainly enriched at 30 and 50 days after embryo formation.The expression levels of serine acetyltransferase(XP_015865688.1),serine hydroxymethyltransferase(XP_015868935.1),and 3-phosphoshikimate 1-carboxyvinyltransferase(XP_015888682.1),which are involved in amino acid metabolism,showed a continuous increase during embryo development in em-bryo-fertile'Sour jujube',while in embryos of the low-fertility cultivar'Kongfusucui jujube',their expression first decreased and then increased.In particular,serine acetyltransferase(XP_015865688.1)was significantly down-regulated at the peak of abortion(40 days after embryo formation)during the KD4 vs.SD4 period,suggesting that it plays a key role in regulating amino acid accumulation.
