通过分析阿勒泰羊繁殖与非繁殖季节的瘤胃微生物菌群结构差异,探究菌群组成与生殖激素水平及季节性繁殖特性的相关性,为阐明瘤胃微生物在绵羊季节性繁殖调控中的潜在作用提供参考.选择体质量相近、健康状况良好(12±1)月龄的6只阿勒泰母羊,分别于繁殖季节(BS组)和非繁殖季节(N-BS组)采集血液及瘤胃液样品,测定繁殖与非繁殖季节阿勒泰羊的血清生殖激素(FSH、E2、LH、P)浓度,宏基因组测序分析阿勒泰羊瘤胃菌群的组成及相关功能基因的KEGG通路,并对秩和检验筛选的30个差异优势菌属与血清生殖激素水平进行相关性分析.结果表明:(1)阿勒泰羊BS组血清中FSH、E2、LH、P激素水平极显著高于N-BS组(P<0.01).(2)BS组与N-BS组瘤胃菌群在门、属水平上具有明显差异,繁殖季节优势菌属为甲基球菌属(Methylocucumis)、克雷伯氏菌属(Klebsiella)、气球菌属(Aerococcus)和乳酪杆菌属(Lacticaseibacil-lus),其中甲基球菌属(Methylocucumis)与FSH呈正相关,乳酪杆菌属(Lacticaseibacillus)、气球菌属(Aero-coccus)与E2呈正相关,甲基球菌属(Methylocucumis)、气球菌属(Aerococcus)、克雷伯氏菌属(Klebsiella)与LH呈正相关,甲基球菌属(Methylocucumis)与P呈正相关.(3)与N-BS组相比,BS组的功能基因在代谢途径、光合作用、氧化磷酸化等11条KEGG通路中显著富集.综上所述,瘤胃微生物菌群结构与阿勒泰羊的季节性繁殖存在相关性,可能参与调控机体的能量代谢及繁殖过程.

This study aims to clarify the correlation between rumen microbiota composition and repro-ductive hormone levels as well as seasonal reproductive characteristics by analyzing the differences in rumen microbiota community structure of Altay sheep during breeding season(BS)and non-breeding season(N-BS),so as to provide a reference for elucidating the potential role of rumen microbiota in regulating seasonal reproduction in sheep.Six healthy Altay ewes aged(12±1)months with similar body mass were selected.Blood and rumen fluid samples were collected from these ewes in the BS and N-BS,respectively.Serum reproductive hormone concentrations(FSH,E2,LH,P)were measured,and metagenomic sequencing was performed to analyze the composition of the rumen microbiota and the KEGG pathways of related functional genes.Correlation analysis was conducted between serum re-productive hormone levels and 30 differential dominant genera at the genus level screened by rank sum test.The results showed that:(1)The concentrations of FSH,E2,LH,and P in the BS group were extremely significantly higher than those in the N-BS group(P<0.01).(2)There were significant differences in rumen microbiota at the phylum and genus levels between the BS and N-BS groups.The dominant genera in the breeding season were Methylocucumis,Klebsiella,Aerococcus,and Lactica-seibacillus.Among them,Methylocucumis was positively correlated with FSH,Lacticaseibacillus and Aerococcus were positively correlated with E2,Methylocucumis,Aerococcus,and Klebsiella were positively correlated with LH,and Methylocucumis was positively correlated with P.(3)Compared with the N-BS group,the functional genes in the BS group were significantly enriched in 11 KEGG pathways,including metabolic pathways,photosynthesis,and oxidative phosphorylation.In conclu-sion,rumen microbiota community structure is correlated with the seasonal reproduction of Altay sheep and may participate in regulating energy metabolism and reproductive processes.
