益生菌功能发挥的物质基础与研究新范式OA

Material Basis and Emerging Research Paradigms of Probiotic Functions

益生菌在调节宿主代谢、免疫稳态及肠道微生态平衡中的作用备受关注,但其功能发挥的物质基础与研究范式仍在持续演进.该综述系统梳理近年益生菌研究进展,从"物质基础"与"研究范式"两条主线概述关键机制与证据框架.在物质基础方面,归纳益生菌表面结构成分(黏附素、菌毛、胞外多糖及细胞壁微生物相关分子模式)与宿主受体轴的互作规律,阐释其在定植、免疫调节与信号转导中的作用;同时总结短链脂肪酸、胆汁酸及其代谢物、维生素、多胺及神经活性化合物等代谢产物对宿主代谢与信号通路的调控,并从生态学视角讨论交叉互养、生态位占据与定植抗性维持肠道稳态的机制.在研究范式方面,概述单细胞技术、多组学整合、器官芯片、人源化动物模型与临床研究等多模态方法对功能解析与因果验证的推动作用,并结合精准营养与个体化医学趋势提出未来应用方向与研究重点.

Probiotics have garnered extensive attention for their roles in regulating host metabolism,immune homeostasis,and gut ecological balance;however,the material basis underlying these functions and the prevailing research paradigms continue to evolve.This review summarizes recent progress in probiotic research from two perspectives:material basis and research paradigms.With respect to material basis,it outlines how probiotic surface-associated components(including adhesins,pili,exopolysaccharides,and cell wall microbe-associated molecular patterns)interact with host receptor axes and elucidates their roles in colonization,immunomodulation,and signal transduction.It also summarizes the roles of probiotic-derived metabolites,including short-chain fatty acids,bile acids and their metabolites,vitamins,polyamines,and neuroactive compounds,in regulating host metabolism and signaling pathways.In addition,it discusses how cross-feeding,niche occupation,and colonization resistance contribute to maintaining gut homeostasis.Regarding research paradigms,it reviews and highlights how emerging multimodal methods such as single-cell technologies,multi-omics integration,organ-on-a-chip platforms,humanized animal models,and clinical studies advance functional dissection and causal verification.Finally,in the context of precision nutrition and individualized medicine,it outlines future application directions and research priorities for probiotics.
