试验旨在筛选新疆本土具备饲用开发潜力的益生芽孢杆菌.以新疆石河子地区健康荷斯坦奶牛新鲜粪便为样品,经热处理富集芽孢菌后分离纯化,通过形态学观察、16S rRNA基因测序完成菌种鉴定,系统评价菌株的益生特性与生物安全性.结果显示,分离获得1株地衣芽孢杆菌NXJ-1,该菌株为γ溶血;其生长性能优异,接种2 h后进入对数生长期,最适生长pH值为6.0;在人工胃液、肠液中孵育3.0 h后,存活率分别达72.00%、84.10%;24 h自凝率为40.12%.抗生素敏感性初筛显示,该菌株对20种畜禽常用抗生素均有不同程度的抑菌作用,其中16种抗生素抑菌圈直径>20.0 mm,未发现明显耐药表型.小鼠安全性试验表明,各剂量组小鼠均无异常,菌株对小鼠生长性能无不良影响;其中中剂量组可以极显著提高血清总蛋白含量(P<0.01),对肾脏功能核心指标无显著影响.研究表明,地衣芽孢杆菌NXJ-1益生特性优异、生物安全性可靠,具有良好的饲用开发潜力,可为新疆本土反刍动物养殖中抗生素的替代提供参考.

This study aimed to screen probiotic Bacillus strains with potential for feed development indigenous to Xinjiang.Fresh feces from healthy Holstein dairy cattle in Shihezi,Xinjiang were used as samples.Spore-forming bacteria were enriched by heat treatment,followed by isolation and purification.Strain identification was accomplished through morphological observation and 16S rRNA gene sequencing.The probiotic properties and biosafety of the strain were systematically evaluated.The results showed that one strain of Bacillus licheniformis NXJ-1 was isolated,exhibiting γ-hemolysis.It demonstrated excellent growth performance,entering the logarithmic growth phase two hours after inoculation,with an optimal growth pH of 6.0.After three hours of incubation in simulated gastric fluid and intestinal fluid,the viable cell survival rates reached 72.00%and 84.10%,respectively.The 24-hour auto-aggregation rate was 40.12%.Preliminary antibiotic susceptibility screening revealed that the strain exhibited varying degrees of inhibitory response to 20 commonly used veterinary antibiotics,with inhibition zone diameters>20.0 mm for 16 antibiotics,and no significant drug resistance phenotype was observed.Mouse safety tests confirmed that mice in all dosage groups showed no abnormalities,and the strain had no adverse effects on mice growth performance.Notably,the total serum protein content in the medium-dose group was extremely increased(P<0.01),with no significant effects on core indicators of kidney function.The study shows that Bacillus licheniformis NXJ-1 possesses excellent probiotic properties and reliable biosafety,demonstrating good potential for forage development,which may provide a reference for antibiotic alternatives in indigenous ruminant farming in Xinjiang.
