姜黄素和咖啡酸对鼠伤寒沙门氏菌感染文昌鸡的保护作用OA
Protective effects of curcumin and caffeic acid on Wenchang chickens infected with Salmonella typhimurium
试验旨在探究姜黄素(CUR)和咖啡酸(CA)对鼠伤寒沙门氏菌(ST)感染文昌鸡生长性能、抗氧化能力、肠道屏障功能及炎症反应的影响.选取180只1日龄雄性文昌鸡,随机分为6组,分别为对照组(CON组)、ST攻毒组(ST组)、ST+5 mg/kg环丙沙星组(ST+CIP组)、ST+200 mg/kg CUR组(ST+CUR组)、ST+100 mg/kg CA组(ST+CA组)、ST+200 mg/kg CUR+100 mg/kg CA组(ST+CUR+CA组),每组3个重复,每个重复10只.试验期21 d.结果显示,与ST组相比,ST+CUR组、ST+CA组和ST+CUR+CA组15~21 d平均日增重(ADG)显著提高(P<0.05),料重比(F/G)显著降低(P<0.05).攻毒后第4天和第11天,与ST组相比,ST+CUR组、ST+CA组和ST+CUR+CA组绒毛高度、绒隐比、杯状细胞数量显著提高(P<0.05),回肠紧密连接蛋白和黏蛋白2(MUC2)的表达显著上调(P<0.05),白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的表达显著下调(P<0.05).ST+CUR组和ST+CA组攻毒后第4天超氧化物歧化酶(SOD)活性显著提高(P<0.05),ST+CUR 组、ST+CA 组和 ST+CUR+CA 组攻毒后第 4 天环氧合酶-2(COX-2)的表达量和攻毒后第11天的丙二醛(MDA)含量显著降低(P<0.05).研究表明,CUR和CA单独及其联用均可通过增强抗氧化能力、降低肠道炎症和改善肠道屏障功能,有效缓解ST攻毒导致的文昌鸡生长抑制和肠道损伤,二者联用在感染早期改善肠道黏膜屏障功能和肠道炎症反应方面效果较好.
The experiment aimed to investigate the effects of curcumin(CUR)and caffeic acid(CA)on the growth performance,antioxidant capacity,intestinal barrier function,and inflammatory response of Wenchang chickens infected with Salmonella typhimurium(ST).A total of 180 male Wenchang chickens aged one day were randomly divided into six groups,namely control group(group CON),ST challenge group(group ST),ST+5 mg/kg ciprofloxacin group(group ST+CIP),ST+200 mg/kg CUR group(group ST+CUR),ST+100 mg/kg CA group(group ST+CA),ST+200 mg/kg CUR+100 mg/kg CA group(group ST+CUR+CA),with three replicates in each group and 10 chickens in each replicate.The test period lasted for 21 days.The results showed that compared with the group ST,the average daily gain(ADG)from 15 to 21 days in groups ST+CUR,ST+CA,and ST+CUR+CA was significantly increased(P<0.05),the feed to gain ratio(F/G)was significantly decreased(P<0.05).On the 4th and 11th day after toxin attack,compared with the group ST,the villus height,villus height to crypt depth ratio,goblet cell number in groups ST+CUR,ST+CA,and ST+CUR+CA were significantly increased(P<0.05).The expression of tight junction protein and mucin 2(MUC2)in the ileum were significantly upregulated(P<0.05).The expression of interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)were significantly downregulated(P<0.05).The activity of superoxide dismutase(SOD)on the 4th day after toxin attack was significantly increased in the groups ST+CUR and ST+CA than in the group ST(P<0.05).The expression of cyclooxygenase-2(COX-2)on the 4th day after toxin attack and the content of malondialdehyde(MDA)on the 11th day after toxin attack were significantly reduced in the groups ST+CUR,ST+CA,and ST+CUR+CA(P<0.05).The study shows that CUR,CA and their combination can effectively alleviate the growth inhibition and intestinal injury of Wenchang chickens caused by ST toxin attack by enhancing antioxidant capacity,reducing intestinal inflammation and improving intestinal barrier function,the combination of the two has a better effect on improving intestinal mucosal barrier function and intestinal inflammatory response in the early stage of infection.
