Effects of chlorogenic acid substances on in vitro ruminal fermentation characteristics and microbiota structure in beef cattle

试验旨在探究绿原酸类物质对西门塔尔肉牛瘤胃发酵特性及微生物群落结构的影响.选取3头西门塔尔肉牛采集瘤胃液.对照组(CK组)每瓶加入150 mL混合培养液,绿原酸类物质组(CA组)每瓶在2 g绿原酸类物质基础上加入150 mL混合培养液,每组设置4个重复.在发酵72 h后测定总产气量、pH值、微生物多样性及氨态氮(NH3-N)、微生物蛋白(MCP)和挥发性脂肪酸(VFA)含量.结果显示,与CK组相比,CA组总产气量及MCP、乙酸、丁酸含量极显著提高(P<0.01),pH值极显著降低(P<0.01),NH3-N含量显著降低(P<0.05).CA 组 Chao1 指数、Observed OTUs 指数、Shannon 指数和 Simpson 指数均极显著降低(P<0.01).在门水平上,CA 组变形菌门相对丰度显著升高(P<0.05),放线菌门相对丰度极显著升高(P<0.01),拟杆菌门和帕斯特菌门相对丰度极显著降低(P<0.01);在属水平上,假丁酸弧菌属、普雷沃氏菌属和丁酸弧菌属等产酸菌相对丰度极显著升高(P<0.01),理研菌科RC9肠道群和琥珀酸单胞菌属相对丰度极显著降低(P<0.01).研究表明,绿原酸类物质可以提高瘤胃微生物产酸菌相对丰度,并通过定向调控微生物群落结构改善瘤胃发酵特性.

The study aimed to investigate the effects of chlorogenic acid substances on the ruminal fermentation characteristics and microbiota structure in Simmental beef cattle.Three Simmental beef cattle were selected for rumen fluid collection.The control group(group CK)received 150 mL of mixed culture fluid per bottle,while the chlorogenic acid substances group(group CA)received 150 mL of mixed culture fluid supplemented with 2 g of chlorogenic acid substances per bottle,with four replicates per group.After 72 hours of fermentation,total gas production,pH value,microbial diversity,and the concentrations of ammonia nitrogen(NH3-N),microbial protein(MCP),and volatile fatty acids(VFA)were measured.The results showed that compared with the group CK,the group CA exhibited extremely increases in total gas production,and concentrations of MCP,acetate and butyrate(P<0.01),an extremely decrease in pH value(P<0.01),and a significant decrease in NH3-N concentration(P<0.05).The Chao1 index,Observed OTUs index,Shannon index,and Simpson index in group CA were extremely decreased(P<0.01).At the phylum level,the relative abundance of Proteobacteria in group CA was significantly increased(P<0.05),the relative abundance of Actinobacteria was extremely increased(P<0.01),while the relative abundances of Bacteroidetes and Pasteurellaceae were extremely decreased(P<0.01).At the genus level,the relative abundances of acid-producing bacteria such as Pseudobutyrivibrio,Prevotella,and Butyrivibrio were extremely increased(P<0.01),while the relative abundances of Rikenellaceae RC9 gut group and Succinimonas were extremely decreased(P<0.01).The study shows that chlorogenic acid substances can increase the relative abundance of acid-producing bacteria in the rumen microbiota and improve ruminal fermentation characteristics through targeted regulation of the microbial community structure.
