Influence of Melatonin on Physiological Characteristics of Flue-Cured Tobacco Seedlings under Low Nitrogen Stress

为明确外源褪黑素对低氮胁迫下烤烟幼苗生理特性的影响,本试验以烤烟品种 9A05 和 9K30 为试材,设置 CK(对照:4 mmol·L-1 NO-3+0 μmol·L-1 褪黑素)、CKM(4 mmol·L-1 NO-3+100 μmol·L-1 褪黑素)、LN(0.4 mmol·L-1 NO-3+0 μmol·L-1褪黑素)、LNM(0.4 mmol·L-1 NO-3+100 μmol·L-1褪黑素)共 4 个处理,研究叶面喷施褪黑素对低氮胁迫下烤烟幼苗形态特性、光合性能、渗透调节和氮素利用的影响.结果表明:(1)喷施褪黑素可以缓解低氮胁迫对烤烟幼苗生长的抑制,与 LN 相比,LNM 处理增加 9A05 的株高、茎围、叶面积、地上部鲜重和根系鲜重且多达显著水平,增幅分别为 40.61%、52.83%、17.86%、27.58%和 34.06%.(2)喷施褪黑素可以提高低氮胁迫下烤烟幼苗的 SPAD 值,与 LN 相比,LNM 处理显著增加 9K30 的 SPAD 值,增幅为15.53%,对 9A05 也有一定的促进作用,增幅为 2.68%.(3)喷施褪黑素可以提升低氮胁迫下烤烟幼苗的光合特性,与 LN 相比,LNM 处理提高品种 9A05 和 9K30 的净光合速率(Pn),增幅分别为 17.12%和 23.37%.(4)喷施褪黑素可以提高低氮胁迫下烤烟幼苗的氮代谢,与 LN 相比,LNM 处理提高幼苗叶片和根系中的硝态氮含量,提高叶片的硝酸还原酶活性.(5)喷施褪黑素可以影响低氮胁迫下烤烟幼苗的渗透调节物质含量,可溶性蛋白和脯氨酸含量下降,可溶性糖含量得到提升.(6)对不同处理下两品种 18 个生理指标进行相关性分析和主成分分析,继而进行综合得分排序,结果为:2 个品种的 LNM 处理均优于 LN 处理.综之,施加褪黑素可以缓解低氮胁迫对烤烟幼苗的影响,促进烤烟幼苗生长.

To explore the effect of melatonin on physiological characteristics of flue-cured tobacco seed-lings under low nitrogen stress,the experiment was carried out by using flue-cured tobacco varieties 9A05 and 9K30 as experimental materials,and setting four treatments of CK(the control,4 mmol·L-1 NO-3+0 μmol·L-1 melatonin),CKM(4 mmol·L-1 NO-3+100 μmol·L-1 melatonin),LN(0.4 mmol·L-1 NO-3+0 μmol·L-1 me-latonin)and LNM(0.4 mmol·L-1 NO-3+100 μmol·L-1 melatonin).The effects of foliar spray of melatonin on the morphological characteristics,photosynthetic performance,osmotic regulatory and nitrogen utilization of flue-cured tobacco seedlings under low nitrogen stress were analyzed.The results were as follows.(1)Spraying melatonin could alleviate the inhibition of low nitrogen stress on the growth of flue-cured tobacco seedlings.Compared with LN,the LNM treatment increased the plant height,stem circumference,leaf area,fresh weight of above-ground part and root fresh weight of 9A05 by 40.61%,52.83%,17.86%,27.58%and 34.06%respectively,and most reached significant level.(2)Spraying melatonin could increase the SPAD value of flue-cured tobacco seedlings under low nitrogen stress.Compared with LN,the LNM treatment signifi-cantly increased the SPAD value of 9K30 by 15.53%,and it also had a certain promoting effect on 9A05 with an increase of 2.68%.(3)Spraying melatonin could enhance the photosynthetic characteristics of flue-cured tobacco seedlings under low nitrogen stress.Compared with LN,the LNM treatment increased the net photo-synthetic rate(Pn)of the varieties 9A05 and 9K30 with the increase of 17.12%and 23.37%,respectively.(4)Spraying melatonin could enhance the nitrogen metabolism of flue-cured tobacco seedlings under low ni-trogen stress.Compared with LN,the LNM treatment increased the content of nitrate nitrogen in the leaves and roots of seedlings,and improved the activity of nitrate reductase in the leaves.(5)Spraying melatonin could affect the content of osmotic regulatory substances in flue-cured tobacco seedlings under low nitrogen stress.The contents of soluble protein and proline decreased,while the content of soluble sugar increased.(6)Through correlation analysis and principal component analysis based on the tested 18 physiological indicators of the two varieties,followed by a comprehensive analysis,it was concluded that the LNM treatment was superior to the LN treatment for the two varieties.In conclusion,spraying melatonin could alleviate the low nitrogen stress to flue-cured tobacco seedlings and promote their growth.
