Research on development,performance evaluation and application of online ATP detection equipment

三磷酸腺苷(adenosine triphosphate,ATP)作为细胞的主要能量货币,其含量变化表征微生物的活性水平.但由于在线检测技术手段的缺乏,极大地限制了该指标在相关领域的推广与应用.为此,本研究建立了 ATP 在线检测方法,开发了 ATP在线检测设备,并且分别从设备检测限、稳定性和准确度三个方面评估仪器的性能.随后,将该仪器应用于餐厨废水处理和芽孢杆菌发酵两类生物反应过程,通过测定ATP 数据,分别分析 ATP 浓度与污染物去除效率、菌量的关系.结果表明,该设备相对光子强度(relative light units,RLU)的检测限为 174RLU~2×107 RLU;对不同浓度ATP 标准液进行多次重复测定,检测结果最低相对偏差为 3.30%,说明该设备稳定性较好;对相同样本该检测设备的检测值与酶标仪检测值呈线性关系(R2 为0.970 4),说明该设备的检测准确性高.餐厨废水活性污泥与芽孢杆菌的 ATP 响应规律表明,ATP 浓度可有效表征系统处理效能和微生物代谢活性,ATP 比菌量指标能够提前指示菌群生物量进入活跃期.

Adenosine triphosphate(ATP)serves as the primary energy currency of cells,and changes in its content reflect the activity level of microorganisms.However,the lack of online detection techniques has greatly restricted the popularization and application of this indicator in relevant fields.To address this issue,this study established an online ATP detection method and developed a corresponding online ATP detection device.The performance of the instrument was evaluated in terms of its detection limit,stability and accuracy.Subsequently,this instrument was applied to two categories of biological reaction processes:a kitchen wastewater bioreactor and a Bacillus fermentation process,with ATP data obtained to analyze its correlations with pollutant removal efficiency and bacterial biomass,respectively.The results showed that the photon detection limit of the device was 174RLU~2×107 RLU;repeated measurements of ATP standard solutions at different concentrations yielded a minimum relative deviation of 3.30%,indicating good stability of the device.The detection values of the device for the same samples exhibited a linear relationship with those obtained using a microplate reader(R2=0.970 4),demon-strating high detection accuracy of the device.The ATP response trends of activated sludge in kitchen wastewater and Bacillus revealed that ATP concentration could effectively character-ize the system treatment efficiency and microbial metabolic activity.Moreover,ATP preceded bacterial biomass in indicating the entry of the microbial community into the active growth phase.
