Distribution characteristics of antibiotic resistance genes and mobile genetic elements in typical domestic sewage in Weifang City

[背景]污水环境是抗生素抗性基因(ARGs)富集、扩散与传播的重要场所.可移动遗传元件(MGEs)可介导 ARGs的水平转移.探究污水环境选择压力下二者的关系具有重要的公共卫生学意义. [目的]在潍坊市 3座污水处理厂采集污水样本,检测耐药菌株携带的 ARGs与 MGEs的分布特征,旨在为污水环境中ARGs的传播防控提供科学依据. [方法]采集潍坊市 3座污水处理厂(城北、城西、沙窝)进水口的多份污水样本.采用含阿莫西林、头孢拉定、红霉素、链霉素、万古霉素、氧氟沙星、磺胺嘧啶、四环素 8种单一抗生素的选择性培养基分离、培养耐药菌,通过一代 16s rDNA测序鉴定菌种.采用纸片扩散法(K-B法)测定耐药菌对 6种抗生素的敏感性,参照美国临床和实验室标准协会(CLSI)标准以抑菌圈直径评估耐药表型.采用实时荧光定量 PCR技术检测 7种 ARGs(β-内酰胺类 blaTEM、氨基糖苷类 aac(6')-Ib-cr等)与 2种 MGEs(整合子 intI1和转座子 tnpA-IS4)的相对丰度,以16s rRNA作为内参基因.采用Pearson相关分析计算ARGs与MGEs的相关性. [结果]3座污水处理厂共检出 15种耐药菌,其中中间气单胞菌(检出率 24.39%)和河溪气单胞菌(检出率 19.51%)为共同优势菌种.氧氟沙星和红霉素的抑菌效果显著(部分抑菌圈直径达 32~41 mm),阿莫西林与头孢拉定的抑菌效果较差(抑菌圈直径≤10 mm).本研究共检出4株泛耐药菌株,分别为城北污水处理厂的 3株(抗阿莫西林厦门希瓦氏菌、抗红霉素河溪气单胞菌、抗磺胺嘧啶中间气单胞菌)与沙窝污水处理厂的 1株(抗阿莫西林魏斯腾步假单胞菌).实时荧光定量 PCR结果显示,各污水处理厂代表性耐药菌中检出包括 blaTEM、aa-dA-01、tetG等多种ARGs,以及intI1和tnpA-IS4等MGEs,相对丰度分布于9.226×10-8~1.186×10-3 之间.相关性分析显示,整合子 intI1与 ARGs之间以及转座子 tnpA-IS4与 ARGs之间均呈正相关(r=0.519,P=0.039 4;r=0.507,P=0.031 9). [结论]潍坊市典型生活污水中的优势耐药菌为中间气单胞菌和河溪气单胞菌,耐药菌种类多样,且部分菌株呈现泛耐药表型;其携带的整合子 intI1、转座子 tnpA-IS4等 MGEs可能介导 ARGs的水平转移.

[Background]Sewage environments serve as crucial reservoirs for the enrichment,diffusion,and transmission of antibiotic resistance genes(ARGs).Mobile genetic elements(MGEs)can me-diate horizontal transfer of ARGs.Exploring their relationships under selective pressure in waste-water environments is of significant public health importance. [Objective]To investigate the distribution profiles of ARGs and MGEs among antibiotic resistant bacteria isolated from three municipal wastewater treatment plants(WWTPs)in Weifang City,thereby providing a scientific basis for mitigating ARGs dissemination in sewage environments. [Methods]Influent samples were collected from three WWTPs(Chengbei,Chengxi,and Shawo)in Weifang City.Antibiotic-resistant bacteria were isolated and cultured using selective media supplemented with eight individual antibiotics(amoxicillin,cefradine,erythromycin,streptomycin,vancomycin,ofloxacin,sulfadiazine,and tetracycline).Bacterial identification was performed using Sanger 16s rDNA gene sequencing.Antimicrobial susceptibility to six antibiotics was evaluated using the Kirby-Bauer(K-B)disk diffusion method,with resistance phenotypes determined according to the Clinical and Laboratory Standards Institute(CLSI)guidelines.The relative abundances of seven ARGs[e.g.,β-lactam blaTEM,and aminoglycoside aac(6')-Ib-cr]and two MGEs(integron intI 1 and transposon tnpA-IS4)were quantified via real-time quantitative PCR(qPCR)using the 16s rRNA as an internal reference.Correlations between ARGs and MGEs were evaluated using Pearson correlation analysis. [Results]A total of 15 drug-resistant bacterial species were detected,with Aeromonas media(detection rate:24.39%)and Aeromonas rivipollensis(detection rate:19.51%)identified as the co-dominant species.Ofloxacin and erythromycin demonstrated potent antibacterial efficacy(part inhibition zone diameters of 32-41 mm),whereas amoxicillin and cefradine showed weak efficacy(inhibition zone diameters≤10 mm).Four extensively drug-resistant strains were detected:three from Chengbei WWTP(amoxicillin-resistant Shewanella xiame-nensis,erythromycin-resistant Aeromonas rivipollensis,and sulfadiazine-resistant Aeromonas media)and one from Shawo WWTP(amox-icillin-resistant Pseudomonas weihenstephanensis).qPCR analysis revealed the ubiquitous presence of multiple ARGs(e.g.blaTEM,aadA-01,and tetG)and MGEs(intI1 and tnpA-IS4)across representative strains,with relative abundances ranging from 9.226×10-8 to 1.186×10-3.Correlation analysis demonstrated significant positive correlations between the relative abundance of ARGs and integron intI1(r=0.519,P=0.039 4),and tnpA-IS4(r=0.507,P=0.031 9). [Conclusion]Aeromonas media and Aeromonas rivipollensis constitute the dominant antibiotic-resistant bacterial populations in typical municipal wastewater in Weifang City.The composition of these drug-resistant bacteria is highly diverse,with certain strains exhibiting severe multidrug-resistant phenotypes.Furthermore,MGEs such as integron intI1 and transposon tnpA-IS4 carried by these bacteria play a critical role in mediating the horizontal transfer of ARGs within the sewage environment.
