基于16S rRNA宏基因组技术探究口腔白斑病的微生物群落特点OA
Characterization of the microbial community in oral leukoplakia using 16S rRNA metagenomic sequencing
目的:分析口腔白斑(oral leukoplakia,OLK)的微生物群落特点,以及伴异常增生和不伴异常增生白斑的微生物分布的差异,探讨口腔微生物群落在白斑癌变进程中可能发挥的作用.方法:分别纳入经病理确认上皮异常增生和无上皮异常增生的口腔白斑患者,收集病损处黏膜拭子,采用 16S rRNA 宏基因组技术对收集的口腔样本进行微生物测序分析,以健康人群口腔样本作为对照,比较各组间微生物多样性和门水平、属水平上的差异.结果:与健康对照组相比,白斑病损处微生物多样性显著降低(Chao 指数 P<0.001),且微生物组成存在明显差异.在菌群组成方面,白斑病损处 Neisseria、Prevotella_7、Campylobacter 等菌属显著富集,而 Haemophilus 和 Streptococcus 则明显减少.特别值得注意的是,伴上皮异常增生的白斑和无异常增生的白斑菌群组成存在不同,前者较后者表现出 Fuso-bacterium、Stenotrophomonas、Aggregatibacter 和 Porphyromonas 的显著增加.结论:本研究揭示了口腔白斑病损处微生物群落的变化特点,为深入探究口腔白斑发病及癌变机制提供了新线索,也为口腔白斑的癌变预防治疗提供了新思路.
Objective:To analyze the characteristics of the microbial community in oral leukoplakia(OLK)and the differences in microbial distribution between OLK with and without epithelial dysplasia,thereby exploring the potential role of the oral microbiota in the malignant transformation of OLK.Methods:Patients with histologically confirmed OLK,both with and without epithelial dysplasia,were enrolled.Mucosal swabs were collected from the lesions.Microbial sequencing and anal-ysis of the collected oral samples were performed using 16S rRNA metagenomic technology.Oral samples from healthy indi-viduals served as controls.Differences in microbial diversity and composition at the phylum and genus levels were compared among the groups.Results:Compared with the healthy control group,the microbial diversity in OLK lesions was significantly reduced(Chao index,P<0.001),and the microbial composition was markedly different.In terms of microbiota composition,genera such as Neisseria,Prevotella_7,and Campylobacter were significantly enriched in OLK lesions,while Haemophilus and Streptococcus were significantly reduced.Of particular note,the microbial composition differed between OLK with dysplasia and OLK without dysplasia.The former showed a significant increase in Fusobacterium,Stenotrophomonas,Aggregatibacter,and Porphyromonas compared to the latter.Conclusion:This study reveals the characteristics of microbial community altera-tions in oral leukoplakia lesions.It provides new clues for in-depth research into the pathogenesis and malignant transforma-tion mechanisms of OLK,and also offers new insights for the prevention and treatment of OLK carcinogenesis.
