拉曼光谱激光诱导牙本质荧光背景的物质来源及过氧化物漂白剂对其影响机制的研究OA
Study on the source of Raman spectral laser-induced dentin fluorescence background and its influence mecha-nism by peroxide bleaching agents
目的:利用拉曼光谱等技术分析过氧化物漂白剂处理后牙本质激光诱导荧光背景的变化规律,明确荧光背景变化的物质来源.方法:将 54 个牙本质切片样本随机分成 3 组(n=18),分别放入 10%中性过氧化脲(car-bamide peroxide,CP)、40%中性过氧化氢(hydrogen peroxide,HP)和去离子水对照组中,连续作用 24 h.利用拉曼光谱仪和红外光谱仪分别检测不同试剂处理前后的拉曼光谱和红外光谱,扫描电镜观察各组牙本质表面形貌特征.提取各组非胶原蛋白成分,进行凝胶电泳和质谱分析.结果:拉曼光谱结果显示牙本质的无机质中的磷酸根峰值保持相对稳定,而激光诱导荧光背景强度在过氧化脲和过氧化氢处理后显著降低(P<0.05).红外光谱结果显示过氧化物漂白剂处理导致胶原蛋白的酰胺峰呈现明显下降趋势(P<0.05).扫描电镜观察到牙本质胶原纤维形态完整,其断裂面形态在处理过程中并未有明显改变.SDS-PAGE 结果表明非胶原蛋白的含量在漂白组显著减少.质谱分析结果进一步表明多种与细胞外基质重塑、蛋白降解相关的非胶原蛋白发生显著差异表达.结论:牙本质非胶原蛋白是拉曼光谱激光诱导荧光背景的主要物质来源.过氧化物漂白剂通过氧化修饰与降解非胶原蛋白使荧光背景强度显著下降,且过氧化氢的氧化效能强于过氧化脲,为深入理解牙齿漂白分子机制提供了实验依据.
Objective:To analyze changes in laser-induced fluorescence background following peroxide bleaching a-gent treatment using Raman spectroscopy and related techniques,identify the source of fluorescence variations.Methods:Fif-ty-four dentin sections were randomly assigned to three groups:10%neutral urea peroxide,40%neutral hydrogen peroxide,and deionized water control(n=18 each),with 24-hour reagent exposure.Raman and infrared spectroscopy were performed before and after treatment.Scanning electron microscopy examined dentin surface morphology,and non-collagen proteins were extracted for gel electrophoresis and mass spectrometry analysis.Results:Raman spectroscopy showed stable phosphate peaks while laser-induced fluorescence background intensity significantly decreased in both bleaching groups.Infrared spectroscopy demonstrated marked decline in collagen amide peaks.Scanning electron microscopy confirmed intact collagen fiber morphol-ogy.SDS-PAGE revealed marked reduction in non-collagen protein content,and mass spectrometry identified significant dif-ferential expression of non-collagen proteins associated with extracellular matrix remodeling and protein degradation.Conclu-sion:Dentin non-collagen proteins,rather than collagen,are the primary source of laser-induced fluorescence background;their oxidative degradation by peroxide bleaching agents directly reduces this background,providing molecular-level insight into tooth bleaching mechanisms.
