基于网络药理学的灵芝调控肿瘤微环境治疗胰腺癌的机制OA
The Mechanism of Treatment of Ganoderma lingzhi on Pancreatic Cancer by Regulating and Controlling Tumor Microenvironment Based on Network Pharmacology
为探讨灵芝Ganoderma lingzhi通过调控肿瘤微环境治疗胰腺癌(PC)的潜在分子机制,通过整合生物信息学与网络药理学方法,系统分析了灵芝活性成分的作用靶点及通路.通过TCMSP数据库及文献筛选获得包括灵芝酚A、灵芝酸V1、赤芝孢子内酯B等在内的灵芝活性成分39种,并利用SwissTarget Prediction数据库预测其潜在靶点;结合ESTIMATE算法评估TCGA数据集中胰腺癌样本的免疫与基质评分,运用WGCNA识别肿瘤微环境相关基因模块,进而与Drugbank、GeneCards、DisGeNET数据库中的疾病靶点及灵芝活性靶点取交集,得到治疗胰腺癌的潜在核心靶点VEGFA、KDR、NTRK1等53个.通过构建蛋白相互作用网络、GO功能与KEGG通路富集分析,发现这些靶点主要参与炎症反应、信号转导以及Ras、Rapl等信号通路.分子对接验证显示,核心靶点与灵芝活性成分具有稳定的结合能力.初步阐明了灵芝可能通过多成分、多靶点、多通路协同调控肿瘤微环境,从而抑制胰腺癌的发展,为进一步揭示灵芝抗胰腺癌的作用机制提供了理论依据.
To investigate the potential molecular mechanisms by which Ganoderma lingzhi modulates the tumor microenvironment to treat pancreatic cancer(PC),an integrative approach combining bioinformatics and network pharmacology was employed to systematically analyze the targets and pathways of G.lingzhi active compounds.A total of 39 active constituents of G.lingzhi,including Ganoderic acid A,Ganoderic acid V1,and Lingzhi spore lactone B,were identified through the TCMSP database and literature review.Their potential targets were predicted using the SwissTarget Prediction database.The ESTIMATE algorithm was applied to assess immune and stromal scores in pancreatic cancer samples from the TCGA dataset,and weighted gene co-expression network analysis(WGCNA)was utilized to identify gene modules associated with the tumor microenvironment.By intersecting these modules with disease-related targets from DrugBank,GeneCards,and DisGeNET databases and the predicted targets of Ganoderma active compounds,53 potential core targets for pancreatic cancer treatment were identified,including VEGFA,KDR,and NTRK1.Protein-protein interaction network construction,along with Gene Ontology(GO)functional and KEGG pathway enrichment analyses,revealed that these targets are primarily involved in inflammatory responses,signal transduction,and pathways such as Ras and Rap1 signaling.Molecular docking validation demonstrated stable binding affinities between core targets and G.lingzhi active compounds.This study preliminarily elucidates that G.lingzhi may exert anti-pancreatic cancer effects through a multi-component,multi-target,and multi-pathway synergistic regulation of the tumor microenvironment,providing a theoretical foundation for further exploration of its therapeutic mechanisms.
