基于非靶向代谢组学解析不同水稻品种的品质差异OA

Untargeted-metabolomics dissection of quality differences among rice cultivars

为比较相同储藏温度下不同品种稻米中品质相关代谢物的构成差异,采用超高效液相色谱-串联质谱(UPLC-MS/MS)非靶向代谢组学技术,对籼稻桃湘优188(TXY188)和五优 308(WY308)储藏前与恒温条件储藏 90 d后的差异代谢产物进行分析并解析其代谢通路.结果表明,通过 UPLC-MS/MS 共检测出3 055种代谢物.储藏初期,与 WY308相比有239种代谢物在 TXY188中高度积累,丁香亭、香草醛、肉桂酸和松柏醛等代谢物在 TXY188 中显著富集.储藏90 d 后,香草醛、肉桂酸和松柏醛等代谢物含量仍高于 WY308.京都基因与基因组百科全书(KEGG)富集分析结果显示,类黄酮生物合成和苯丙烷代谢是主要代谢通路.本研究从风味代谢物动态变化角度,为稻谷储藏过程中品质的调控提供了数据支持.

To compare the differences in the composition of quality-related metabolites in rice among varieties stored at the same temperature,untargeted metabolomics approach based on ultraperformance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)was used to analyze differential metabolites and elucidate metabolic pathways in the in-dica rice varieties,Taoxiangyou 188(TXY188)and Wuyou 308(WY308),before storage and after 90 d of storage under constant-temperature conditions.The results showed that a total of 3 055 metabolites were detected by UPLC-MS/MS.At the initial stage of storage,239 metabolites were accumulated at higher levels in TXY188 than in WY308,and metabolites such assyringetin,vanillin,cinnamic acid,and coniferaldehyde were significantly enriched in TXY188.After 90 d of storage,the contents of metabolites such as vanillin,cinnamic acid and coniferaldehyde remained higher in TXY188 than in WY308.Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis revealed that flavonoid biosynthesis and phenylpropanoid metabolism were the major metabolic pathways involved.This study provides data support for regulation of grain quality during rice storage from the per-spective of dynamic changes in flavor-related metabolites.
