响应面法优化普通小球藻低温短期保存条件OA

Response Surface Methodology for Optimizing Short-term Cryo-preservation Conditions of Chlorella vulgaris

为优化普通小球藻(Chlorella vulgaris)短期低温保存方法,提高小球藻细胞存活率,为水产经济动物提供优质生物饵料.以普通小球藻为试验藻株,甘油为冷冻保护剂,通过单因素试验确定最适保存温度、藻细胞浓度和保存时间,并采用响应面法进一步优化.结果表明:普通小球藻低温保存时间、藻细胞浓度和保存温度的单因素最适条件分别为12 h,5.0×105/mL和-80℃,上述因素对藻细胞存活率的影响程度为保存时间>保存温度>藻细胞浓度;通过响应面法分析,最佳保存条件为70.179 h,-78.91℃和3.77×105/mL,结合实际操作优化后的保存条件为70 h,-80℃和3.77×105/mL,该条件下普通小球藻复苏24 h后细胞存活率为70.11%.研究结果为小球藻及其他淡水微藻细胞的低温保存提供参考.

This study aimed to optimize the short-term cryopreservation protocol for Chlorella vulgaris and improve the post-resuscitation survival rate of cells,so as to provide high-quality biological feed for economically important aquatic animals.With C.vulgaris used as the test strain and glycerol adopted as the cryoprotectant,the preliminary optimal ranges of three key parameters including cryopreservation temperature,algal cell concentration and preservation duration were determined through single-factor experiments,and the protocol was further optimized by response surface methodology.The results showed that the optimal single-factor conditions for cryopreservation of cells are-80℃,5.0×105 cells/mL,and 12 hours,respectively.The three parameters ranked in descending order of their influence on the survival rate of algal cells as follows:preservation duration>storage temperature>algal cell concentration.Through response surface analysis,the theoretically optimal preservation conditions were identified as a preservation duration of 70.179 h,a storage temperature of-78.91℃,and an algal cell concentration of 3.77×105 cells/mL.Considering the operability in practical experiments,the optimized conditions were adjusted to a preservation duration of 70 h,a storage temperature of-80℃and an algal cell concentration of 3.77×10 ⁵ cells/mL.Under this adjusted protocol,the survival rate of C.vulgaris cells reached 70.11%after 24 h of resuscitation.These findings provided a reliable reference for the establishment of cryopreservation protocols for Chlorella vulgaris and other freshwater microalgal cells.
