表达共轭亚油酸异构酶mcra基因与菌毛亚基工程益生菌的构建及特性OA
Construction and Characteristics of Engineered Probiotics Incorporating the mcra Gene for Conjugated Linoleic Acid Isomerase and Fimbria Subunit Engineering
为开发基于乳酸菌和大肠杆菌的工程益生菌,评估其对肠上皮细胞的黏附和侵袭能力以及对致病性大肠杆菌的竞争黏附与竞争侵袭能力.以干酪乳杆菌ATCC 393和大肠杆菌Nissle 1917为递呈载体,分别表达鼠李糖乳杆菌中编码共轭亚油酸异构酶的mcra基因,以及双歧杆菌黏附性脂蛋白BOPA、产肠毒素大肠杆菌K88菌毛蛋白FaeG和FaeC的融合蛋白,成功构建工程益生菌LC-pPG-mcra和EcN-pPG-BOPA/pBbB8K-FaeC-FaeG(EcN-BFF),并检测工程益生菌对肠上皮细胞Caco-2的黏附与侵袭能力,及其对产肠毒素大肠杆菌K88的竞争黏附与竞争侵袭能力.结果表明:2株工程益生菌对Caco-2细胞均有较高的黏附与侵袭能力,能竞争性抑制K88菌株对Caco-2细胞的感染.成功构建了2株具有良好黏附侵袭能力及抗感染作用的工程益生菌,研究结果为微生态制剂在细菌性肠道疾病防治中的应用奠定基础.
This study aimed to develop engineered probiotics based on Lactobacillus and Escherichia coli and evaluate their adhesion and invasion abilities to intestinal epithelial cells,as well as their competitive inhibition against enterotoxigenic Escherichia coli K88.The mcra gene encoding the linoleate isomerase from Lactobacillus rhamnosus GG and the genes encoding the bifidobacterial adhesive lipoprotein BOPA along with the Enterotoxigenic E.coli K88 fimbrial proteins FaeG and FaeC were successfully expressed by the delivery vectors of Lactobacillus casei ATCC 393 and Escherichia coli Nissle 1917(EcN),respectively.The engineered probiotics were designated as LC-pPG-mcra and EcN-pPG-BOPA/pBbB8K-FaeC-FaeG(EcN-BFF).The adhesion and invasive capabilities of two engineered probiotic strains to Caco-2 intestinal epithelial cells,as well as their competitive adhesion and competitive invasion against ETEC K88,were examined.The results demonstrated that both engineered strains exhibited high adhesion and invasive capabilities to Caco-2 cells.Furthermore,they significantly inhibited the adhesion and subsequent invasion to Caco-2 cells by the pathogenic K88 strain through competitive mechanisms.In conclusion,two engineered probiotics with favorable adhesion,invasion,and anti-infective properties were successfully constructed,these results lays the foundation for the application of microecological agents in the prevention and treatment of bacterial intestinal diseases.
