不同砧木对武鸣沃柑果实成熟期的影响OA

Effects of rootstocks on fruit ripening of Wuming Orah mandarin

为揭示不同砧木对武鸣沃柑成熟期果实品质的影响,明确武鸣沃柑最佳采收期,分别以香橙和枳作为砧木的武鸣沃柑为试材,自果实转色期至成熟期,分8个时期(S1~S8)采样,测定果面色度值、可溶性固形物(total soluble solids,TSS)和可滴定酸(titratable acidity,TA)含量、出汁率以及5个关键时期(S2、S3、S5、S6、S8)的初生代谢物等果实品质指标;采用RNA-Seq分析3个时期(S2、S3、S5)的基因表达差异,并利用qRT-PCR验证关键基因的表达情况.结果显示,S1~S5同一时期,枳砧果实着色更快且果面更红;出汁率提高了0.37%~11.64%,TSS提高了0.73%~1.00%、固酸比提高了0.32~3.41.感官评价试验表明,枳砧果实总体满意度为8.11,显著高于香橙砧果实的总体满意度(7.22).初生代谢组分析表明,枳砧明显提高了果实蔗糖、D-果糖、D-葡萄糖和myo-肌醇含量;而S6~S8同一时期,二者的TA含量、固酸比和感官风味等指标差异不显著.转录组分析共筛选出1 481个差异表达基因,主要富集于碳水化合物代谢、蛋白质磷酸化、转录调控等生物学过程.qRT-PCR证实,果实蔗糖积累关键基因CsSUC1在枳砧果实中优先上调表达.以上结果表明,枳砧嫁接的武鸣沃柑果实最佳采收期为1月上旬,而香橙砧嫁接的沃柑最佳采收期为1月中旬,二者相差约2周;枳砧促进果实成熟的原因主要与加速果实可溶性糖积累有关.

Fragrant citrus(Citrus×junos Siebold ex Tanaka)and Poncirus trifoliata(L.)Raf.graft-ed Wuming Orah mandarin were used to study the effects of different rootstocks on the quality of mature fruits and clarify the optimal harvesting period for Wuming Orah mandarin.Samples of fruit were collected at 8 developmental stages(S1-S8)spanning the color-break period to full maturity to determine the key pa-rameters of quality including the color index of fruit pericarp,the content of total soluble solids(TSS)and titratable acidity(TA),juice yield,and changes in indexes of quality such as primary metabolite profiles at five critical stages.RNA-Seq was used to analyze differences in gene expression at three pivotal stages,and qRT-PCR was used to verify the expression of key genes.Results showed that P.trifoliata-grafted fruits exhibited accelerated pericarp coloration and enhanced redness during S1-S5.The juice yield,TSS,and TSS/TA ratio in P.trifoliata-grafted fruits increased by 0.37%-11.64%,0.73%-1.00%,and 0.32-3.41 compared to that in C.junos-grafted fruits.The results of sensory evaluation showed that the overall prefer-ence score for P.trifoliata-grafted fruits was 8.11,significantly higher than that for C.junos-grafted fruits(7.22).The results of analyzing primary metabolome showed that the content of sucrose,D-fructose,D-glucose,and myo-inositol in P.trifoliata-grafted fruits was significantly increased.There was no significant difference in TA,TSS/TA ratio,and sensory flavor between the two rootstocks during the same period of S6-S8.Transcriptome profiling identified 1 481 differentially expressed genes,predominantly enriched in bio-logical processes including carbohydrate metabolism,protein phosphorylation,and transcriptional regulation.The results of qRT-PCR confirmed the early upregulation of CsSUC1(the key sucrose transporter gene)in P.trifoliata-grafted fruits.It is indicated that the optimal harvesting period for P.trifoliata grafted Wum-ing Orah mandarin occurs in early January,approximately two weeks earlier than that for C.junos grafted fruits(mid-January).The main reason why P.trifoliata rootstock promotes fruit ripening is related to accel-erating the accumulation of soluble sugars in the fruit of Wuming Orah mandarin.
