基于SRAP与AFLP标记的青皮红心柚类遗传关系解析OA
SRAP and AFLP markers-based genetic relationship between green-skinned and red-fleshed pomelos
为解析我国云南河口特色种质青皮红心柚与我国南方主要柚类品种及东南亚部分柚类品种的遗传关系,以32份国内外柚类种质为试验材料,分别筛选出13对SRAP引物与11对AFLP引物进行基因组扩增,结合分子标记技术解析供试种质的遗传多样性与亲缘关系.结果显示,SRAP、AFLP分子标记分别得到125条、91条多态性条带,对应的平均扩增多态性比率为96.40%、95.73%.基于UPGMA聚类分析发现,云南河口青皮红心柚与广西容县沙田柚遗传距离小,云南河口特色种质青皮红心柚云青1号、云青2号与越南青皮红心柚聚为一类,区别于其余16份青柚资源;云南河口青柚513与青柚408遗传距离小,参试18份青皮红心柚资源不存在同物异名、同名异物现象.以上结果表明,SRAP和AFLP标记均可有效用于柚类遗传关系研究,明确了供试青皮红心柚资源与国内外柚资源的群体遗传结构及特色地方种质云青1、2号的遗传背景.
32 pomelos from domestic and international pomelo germplasms were used to study the ge-netic relationship between the green-skinned and red-fleshed pomelos from the distinctive germplasm of Hekou,Yunnan Province,China,the major pomelo varieties in southern China,and some pomelo varieties in Southeast Asia.13 pairs of SRAP primers and 11 pairs of AFLP primers were screened for the amplifica-tion of pomelo genome to analyze the genetic diversity and phylogenetic structure of the pomelos studied.The results showed that the SRAP and AFLP markers produced 125 and 91 polymorphic bands,with an average polymorphism of 96.40%and 95.73%,respectively.The results of cluster analysis with UPGMA showed that genetic distance between green-skinned and red-fleshed pomelos from Hekou Yao Autono-mous County,Honghe Hani and Yi Autonomous Prefecture,Yunnan Province,China and the Shatian pom-elo from Rong County,Guangxi Zhuang Autonomous Region,China was small,while the special germ-plasm with green-skinned and red-fleshed Yunqing No.1 and No.2 from Hekou were grouped with the green-skinned and red-fleshed pomelos from Vietnam,differing from the other 16 green-skinned pomelos.The genetic distance between No.513 and No.408 green-skinned pomelos from Hekou was small.No cases of synonymy or homonymy were found among the 18 green-skinned and red-fleshed pomelos studied.It is indicated that SRAP and AFLP markers can be effectively used for the study of genetic relationships be-tween pomelos to clarify the genetic structure of population in the pomelos from domestic and international pomelo germplasms and the genetic background of pomelo varieties No.1 and No.2 with the local character-istics from Hekou.
