Peanut skin alleviates gentamicin-induced acute kidney injury in rats via regulation of MAPK/PI3K/AKT signalling pathways

目的:探讨花生衣提取物改善急性肾损伤的作用及机制.方法:基于UPLC-Q TOF-MS技术对花生衣提取物及其入血成分进行分析,通过数据库对花生衣提取物入血成分和急性肾损伤(AKI)疾病进行靶点预测,取交集靶点构建"药物-活性成分-靶点-疾病通路"网络,通过GO、KEGG功能富集分析并筛选相关性较高的核心活性成分.通过腹腔注射庆大霉素构建急性肾损伤模型,采用病理组织切片检测肾脏炎症细胞浸润程度,微板法检测肾功能指标、炎症因子含量和肾组织氧化指标;Western blot检测MAPK/PI3K/AKT信号通路的关键蛋白表达水平.结果:基于UPLC-Q TOF-MS技术从花生衣提取物中鉴定出45种化学成分和14种入血成分,构建网络获得131个交集靶点,GO和KEGG富集分析主要涉及MAPK信号通路、PI3K 信 号 通 路、AKT 信 号 通 路.其 中 核 心 成 分 为 Arachidonic Acid、Linoleic Acid、Esculetin、7-Hexadecenoic acid、Daidzein.花生衣提取物可以明显降低AKI大鼠血清中BUN、CRE的含量,降低尿液中UP、LZM酶和NAG含量.减轻TNF-α、IL-1β、IL-6的炎症因子浸润,降低NO、NOS、MDA的水平,提高GSH、SOD、UA的水平.Western blot验证表明,花生衣提取物通过下调MAPK/PI3K/AKT通路相关蛋白磷酸化水平,有效抑制肾小管上皮细胞凋亡.结论:花生衣提取物通过MAPK/PI3K/AKT信号通路缓解庆大霉素诱导的急性肾损伤,为花生衣缓解急性肾损伤的活性成分及药理机制提供了理论支撑.

Objective:To explore the effect and mechanism of peanut skin extract in improving acute kidney injury.Methods:Based on UPLC-Q TOF-MS technology,peanut skin extract and its blood components were analyzed.Target prediction was per-formed on the blood components of peanut skin extract and acute kidney injury disease through a database.The intersection targets were selected to construct a"drug-active ingredient-target-disease pathway"network.GO and KEGG functional enrichment analy-sis were used to analyze the network pathways and screen for core active ingredients with high correlation.An acute kidney injury model was established by intraperitoneal injection of gentamicin.Pathological tissue sections were used to detect the degree of re-nal inflammatory cell infiltration.Microplate method was used to detect renal function indicators,inflammatory factor content,and renal tissue oxidation indicators.Western blot was used to detect the expression levels of key proteins in the MAPK/PI3K/AKT signaling pathway.Results:Based on UPLC-Q TOF-MS technology,45 chemical components and 14 blood entering components were identified from peanut skin extracts.A network was constructed to obtain 131 intersecting targets.GO and KEGG enrich-ment analysis mainly involved the MAPK signaling pathway,PI3K signaling pathway,and AKT signaling pathway.The core components are Arachidonic Acid Linoleic Acid,Esculetin,7-Hexadecenoic acid,and Daidzein.Peanut skin extract can signifi-cantly reduce the levels of BUN and CRE in rat serum induced by gentamicin,as well as decrease the levels of UP,LZM en-zymes,and NAG in urine.Reduce the infiltration of inflammatory factors such as TNF-α,IL-1β,and IL-6,lower the levels of NO,NOS,and MDA,and increase the levels of GSH,SOD,and UA.Western blot validation showed that peanut skin extract effectively inhibited apoptosis of renal tubular epithelial cells by downregulating the phosphorylation levels of key proteins in the MAPK/PI3K/Akt pathway.Conclusion:Peanut skin extract alleviates acute kidney injury induced by gentamicin through the MAPK/PI3K/AKT signaling pathway,providing theoretical support for the active ingredients and pharmacological mechanisms of peanut skin in alleviating acute kidney injury.
