血小板指数与皮肤恶性黑色素瘤的相关性研究OA

目的:研究基因测定的血小板指数与皮肤恶性黑色素瘤(cutaneous malignant melanoma,CMM)之间的关系.方法:从全基因组关联研究(GWAS)提取来自 UK Biobank 数据库的血小板计数(platelet count,PLT)、平均血小板体积(mean platelet volume,MPV)、血小板分布宽度(platelet distribution width,PDW)、血小板压积(thrombocytopenia,PCT)遗传数据和来自FinnGen数据库的CMM遗传数据.使用逆方差加权法(inverse variance weighting,IVW)、MR-Egger回归法、加权中位数方法和加权模式法进行孟德尔随机化分析,并进行敏感性分析;通过体外实验探讨血小板对A375细胞体外表型的影响.结果:PLT对CMM的 IVW 分析示 OR=1.814,95%CI(1.027,3.203),P=0.040;MPV 的 IVW 分析示 OR=0.617,95%CI(0.403,0.945),P=0.026;PDW 的 IVW 分析示 OR=1.148,95%CI(0.674,1.958),P=0.611;PCT 的 IVW分析示OR=1.238,95%CI(0.727,2.110),P=0.432.体外实验证实血小板能促进A375细胞增殖.结论:血小板数量增多和平均体积下降可能是CMM的危险因素;高水平血小板促进A375细胞体外增殖.

Objective:To investigate the association between genetically determined platelet index and cutaneous malignant melanoma(CMM).Methods:Genetic data of platelet count(PLT),mean platelet volume(MPV),platelet distribution width(PDW),and thrombocytopenia(PCT)from the UK Biobank Database and genetic data of CMM from the FinnGen Database were extracted from Genome-wide association studies(GWAS).Inverse variance weighting(IVW),MR-Egger regression,weighted median and weighted pattern methods were used for Mendelian randomization analysis,and sensitivity analysis was per-formed as well.The effect of platelets on the phenotype of A375 cells was studied in vitro.Results:The IVW analysis of PLT for CMM was OR=1.814,95%CI(1.027,3.203),P=0.040;The IVW analysis of MPV for CMM was OR=0.617,95%CI(0.403,0.945),P=0.026;The IVW analysis of PDW was OR=1.148,95%CI(0.674,1.958),P=0.611;The IVW analysis of PCT was OR=1.238,95%CI(0.727,2.110),P=0.432.In vitro experiments confirmed that platelets could promote the pro-liferation of A375.Conclusion:The increase of platelet count and the decrease of mean platelet volume may be the risk factors for CMM.Increased platelets promote A375 cells proliferation in vitro.
