Colorimetric Detection of Ascorbic Acid and Total Antioxidant Capacity Based on CoNi Bimetallic Metal-Organic Framework

采用室温共沉淀法合成了CoNi双金属有机框架材料(CoNi-MOF).得益于Co与Ni之间的双金属协同效应,CoNi-MOF展现出显著优于单金属MOF的类氧化酶(OXD)活性,可催化显色底物3,3',5,5'-四甲基联苯胺(TMB)发生显色反应.当体系中存在抗坏血酸(AA)时,显色反应受到显著抑制,基于此构建了针对AA的比色传感平台.此检测平台对AA的线性检测范围为1~16 μmol/L,检出限(S/N=3)为0.34 μmol/L,并具有良好的选择性与长期稳定性.将此传感器用于新鲜水果及市售饮料的总抗氧化能力(TAC)分析,百香果样品加标回收率为98.17%~101.30%,相对标准偏差(RSD)均低于7%.此检测体系无需外加过氧化氢(H2O2),操作简便、成本低,为食品质量监控与抗氧化能力评估提供了新的分析方法,也为双金属MOF纳米酶在传感分析领域中的应用拓宽了思路.

A CoNi bimetallic metal-organic framework(CoNi-MOF)was synthesized via a room-temperature co-precipitation method.Benefiting from the bimetallic synergistic effect between Co and Ni,CoNi-MOF showed significantly superior oxidase-like(OXD)activity compared with monometallic MOFs,and could catalyze the chromogenic substrate 3,3',5,5'-tetramethylbenzidine(TMB)to produce a color reaction.In the presence of ascorbic acid(AA),the color reaction was markedly inhibited.On this basis,a colorimetric sensing platform for AA detection was constructed.The platform exhibited a linear detection range of 1-16 μmol/L toward AA,with a limit of detection(S/N=3)of 0.34 μmol/L,together with good selectivity and long-term stability.It was successfully applied to analysis of total antioxidant capacity(TAC)in fresh fruits and commercial beverages.The spiked recoveries of passion fruit samples were 98.17%-101.30%,and the relative standard deviations(RSDs)were all below 7%.Without addition of H2O2,this detection system was simple to operate and low in cost,which provided a new analytical method for food quality monitoring and antioxidant capacity evaluation,and expanded new horizons for application of bimetallic MOF nanozymes in sensing analysis.
