目前,单细胞多组学技术已成为生命科学领域的重要前沿技术.相比于单一组学分析,单细胞转录组与蛋白质组的整合分析能够更全面地反映细胞基因表达与功能状态之间的动态关联,为复杂生命过程及疾病机制研究提供了更全面的分子信息.微流控技术凭借其纳升级反应体系及高精度流体操控能力,在单细胞多组学分析中发挥了重要作用.近年来,基于微流控技术,发展了一系列单细胞转录组与蛋白质组等多组学联合分析方法.本文根据微流控技术在各方法中发挥的作用,将转录组与蛋白质组的整合分析策略分为组分分离策略、条形码标记的同步建库策略和空间标记策略,从分析原理、技术特点及其优势与局限性等方面系统综述了基于微流控技术的单细胞转录组与蛋白质组整合分析方法的研究进展,并总结了当前技术发展的关键挑战及该领域未来的发展趋势.

Single-cell multi-omics has become a crucial frontier in life sciences,providing powerful tools for investigating cellular heterogeneity and complex biological processes.Compared with single-omics analysis,the integration of single-cell transcriptomics and proteomics enables a more comprehensive characterization of the dynamic relationship between gene expression and cellular functional states,thereby offering multidimensional molecular information for understanding disease mechanisms.Benefiting from nanoliter-scale reaction systems and precise fluid manipulation,microfluidic technology has played an essential role in the development of integrated single-cell multi-omics analysis.In recent years,a series of microfluidics-based strategies have been developed for simultaneous analysis of single-cell transcriptomes and proteomes.According to the roles of microfluidic technologies in these approaches,the integration strategies can be categorized into three types:component separation strategies,barcode-assisted synchronous library construction strategies,and spatial labeling strategies.This review summarizes recent advances in microfluidics-based integrated single-cell transcriptomic and proteomic analysis from the perspectives of analytical principles,technical characteristics,as well as their advantages and limitations.Finally,the key technical challenges and future development trends in this field are discussed.
