目的 探讨空气细颗粒物(fine particulate matter,PM2.5)对人支气管上皮细胞上皮间质转化(epithelial-mesenchymal transition,EMT)相关分子表达的影响及其时间和剂量效应.方法 采用人支气管上皮细胞系Beas-2b,分别给予不同浓度PM2.5(0、25、50、100 μg/ml)处理,暴露24、48、72 h后收集细胞.实时荧光定量聚合酶链式反应(qRT-PCR)检测E-cadherin、Vimentin、Snail2、ZEB1、ZEB2及MMP-9的mRNA表达.蛋白免疫印迹法检测E-cadherin、Vimentin的蛋白水平.结果 与对照组相比,PM2.5暴露显著降低Beas-2b细胞E-cadherin mRNA表达,且剂量主效应显著(P<0.000 1);Vimentin mRNA表达未见显著改变.蛋白水平验证显示,PM2.5暴露24 h后E-cadherin在50、100 μg/ml暴露组显著下降,而Vimentin蛋白表达无明显变化,与转录水平结果一致.Snail2 mRNA表达存在显著PM2.5暴露剂量和暴露时间交互作用[F(6,24)=7.582,P=0.000 1];简单效应分析显示,PM2.5暴露24 h后仅100 μg/ml暴露组Snail2 mRNA显著升高,暴露48 h和72 h后25、50、100 μg/ml暴露组Snail2 mRNA均显著升高.MMP-9 mRNA表达无显著暴露剂量与时间交互作用,但暴露剂量主效应[(F(3,24)=16.08,P<0.000 1]和暴露时间主效应[F(2,24)=6.979,P=0.004 1]均显著;与对照组相比,25、50、100 μg/ml暴露组总体表达水平均显著升高,且在PM2.5暴露72 h组总体水平高于暴露24 h和48 h组.ZEB1、ZEB2 mRNA未见显著改变.结论 PM2.5诱导支气管上皮细胞上皮特征减弱,该过程可能与Snail2和MMP-9上调有关.

OBJECTIVE To investigate the effects of ambient fine particulate matter(PM2.5)on epithelial-mesenchymal transition(EMT)-related molecules in human bronchial epithelial cells and to define the time-and concentration-dependent pattern of these effects.METHODS Beas-2b cells were exposed to PM2.5 at 0,25,50,100 μg/ml and collected at 24,48,and 72 h.Quantitative real-time PCR(qRT-PCR)was performed to detect the mRNA levels of E-cadherin,Vimentin,Snail2,ZEB1,ZEB2,and MMP-9.Western-blotting was used to detect the protein levels of E-cadherin and Vimentin.RESULTS Compared with the control group,PM2.5 significantly downregulated E-cadherin mRNA in Beas-2b cells,with a significant main effect of concentration(P<0.000 1),whereas Vimentin mRNA levels showed no significant change.At the protein level,E-cadherin was significantly decreased after 24 h of PM2.5 exposure in the 50 and 100 μg/ml groups compared with controls,while Vimentin protein expression remained unchanged,consistent with the transcriptional findings.For Snail2 mRNA,a significant interaction between PM2.5 concentration and exposure time was observed[F(6,24)=7.582,P=0.000 1].Simple effects analysis showed that after 24 h of PM2.5 exposure,Snail2 mRNA was significantly increased only in the 100 μg/ml PM2.5-treated group,whereas after 48 and 72 h of PM2.5 exposure,significant increases were observed in all PM2.5-treated groups(25,50,and 100 μg/ml).No significant interaction between concentration and exposure time was found for MMP-9 mRNA;however,both the main effect of concentration[F(3,24)=16.08,P<0.000 1]and the main effect of exposure time[F(2,24)=6.979,P=0.004 1]were significant.Compared with the control group,the overall MMP-9 mRNA was significantly increased in all PM2.5-treated groups(25,50,and 100 μg/ml),and the overall MMP-9 mRNA level at 72 h of PM2.5 exposure was higher than those at 24 and 48 h.No significant changes were observed in ZEB1 and ZEB2 mRNA levels.CONCLUSION PM2.5 exposure attenuates the epithelial phenotype of human bronchial epithelial cells,and the effect may be mediated,at least in part,by the upregulation of Snail2 and MMP-9.
