针对传统阳极溶出伏安法(anodic stripping voltammetry,ASV)在粮食检测中前处理流程复杂、耗时长,难以满足现场快速检测需求等问题,建立并优化基于ASV快速测定粮食中镉(Cd)和铅(Pb)含量的检测体系.以乙酸铵+氯化铵+汞为底液,体积分数20%的硝酸为提取液,通过涡旋振荡-离心作用简化前处理流程,系统优化底液、提取液、提取时间等关键参数;采用空白基质匹配标准曲线法消除基质干扰,以电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry,ICP-MS)为参比,进行方法比对验证.结果表明,Cd和Pb在0~500 μg/L范围内线性关系良好,糙米中Cd,Pb检出限分别为 0.005,0.006 mg/kg,玉米中分别为 0.011,0.012 mg/kg;平均回收率为 95.3%~102.3%,变异系数为3.16%~10.20%;与ICP-MS法对比,t检验无显著性差异(P>0.05);优化后,样品前处理与测定时长仅需10 min.研究为主粮的现场快速安全检测提供可靠的技术参考.

To address the issues of complex pretreatment procedures,time-consuming operations,and difficulties in meeting the requirements for on-site rapid detection of cadmium(Cd)and lead(Pb)in grains using traditional anodic stripping voltammetry(ASV),an analytical system based on ASV for the rapid determination of Cd and Pb in grains was established and optimized.Ammonium acetate+ammonium chloride+mercury was used as the supporting electrolyte,and 20%(V/V)nitric acid was employed as the extraction solution.The pretreatment process was simplified through vortex oscillation and centrifugation.Key parameters including the supporting electrolyte,extraction solution,and extraction time were systematically optimized.Matrix-matched external calibration was adopted to eliminate matrix effects,and the method was validated by comparison with inductively coupled plasma mass spectrometry(ICP-MS)as the reference method.The results showed that Cd and Pb exhibited good linearity in the concentration range of 0-500 μg/L.The limits of detection(LOD)for Cd and Pb in brown rice were 0.005,0.006 mg/kg,respectively,and those in corn were 0.011,0.012 mg/kg,respectively.The average recoveries ranged from 95.3%to 102.3%,with coefficients of variation(CV)between 3.16%and 10.20%.No significant difference was observed compared with the ICP-MS method by t-test(P>0.05).After optimization,the total time required for sample pretreatment and measurement was only 10 min.This study provides a reliable technical reference for the on-site rapid safety detection of staple grains.
