[目的]探究卷烟纸非水溶黄斑成因.[方法]采用液相色谱-二极管阵列检测器技术,对非水溶黄斑中黄色物质进行定性与定量分析,同时结合萃取优化和溶剂淋洗实验分析其来源.[结果]卷烟纸中非水溶性黄斑的黄色物质溶于乙醇等有机溶剂,不溶于水,在日光照射下颜色会减退,可通过斑点水溶性测试和光照进行辨别.选择乙醇为萃取溶剂,对卷烟纸采用涡旋萃取,对烟草物料采用基质破碎萃取,可将脂溶性黄色色素萃取完全.结合二维吸收光谱图中色素的保留时间和吸收光谱轮廓,定性检测非水溶黄斑卷烟纸中的黄色物质为叶黄素和 β-胡萝卜素;8 个非水溶性黄斑卷烟纸中叶黄素和 β-胡萝卜素含量分别为 1.07~7.00 μg/g 和0.64~13.37 μg/g,酯类溶剂污染形成的卷烟纸油性斑点中也检出少量叶黄素和 β-胡萝卜素.叶黄素和 β-胡萝卜素主要来源于配方烟丝,不同物料中其含量排序为叶丝>薄片>梗丝,经二氯甲烷淋洗后,薄片中脂溶性黄色素的可溶出量最大,梗丝中色素的可溶出比例最高.[结论]本研究建立的测定方法可用于卷烟纸非水溶性黄斑的鉴别及来源识别,可为黄斑烟支的科学防控提供参考.

[Objective]To investigate the material basis and causes of non-water-soluble yellow spots on cigarette paper.[Methods]Liquid chromatography coupled with a diode array detector(LC-DAD)was used to qualitatively and quantitatively analyze the yellow substances in non-water-soluble yellow spots,and their sources were investigated through the determination of yellow substances in different tobacco materials combined with solvent leaching experiments.[Results]The result showed that the yellow substances in the non-water-soluble spots were soluble in organic solvents such as ethanol but insoluble in water,and their color faded upon sunlight exposure.Such spots can be distinguished by testing their water solubility and light fading behavior.With ethanol as the extraction solvent,vortex extraction was applied to cigarette paper and matrix disruption extraction to tobacco materials,achieving complete extraction of the lipophilic yellow pigments.Based on their retention times and absorption spectral profiles obtained by two-dimensional absorption spectroscopy,the yellow substances in the non-water-soluble spots were identified as lutein and β-carotene.The contents of lutein and β-carotene in eight cigarette paper samples with non-water-soluble yellow spots were determined to be in the range of 1.07~7.00 μg/g and 0.64~13.37 μg/g,respectively.Trace amounts of lutein and β-carotene were also detected in oily spots on cigarette paper caused by ester solvent contamination.Lutein and β-carotene mainly originated from the blended cut tobacco,and their contents in different materials followed the order of cut tobacco>reconstituted tobacco sheet>cut stem.Dichloromethane leaching experiments showed that among these materials,reconstituted tobacco sheet released the largest amount of lipophilic yellow pigments,whereas cut stem exhibited the highest leachable proportion.[Conclusion]The established method can be used for the identification and source tracing of non-water-soluble yellow spots on cigarette paper,providing a reference for the scientific prevention and control of spotted cigarettes.
