基于网络药理学与分子对接探讨生血宝合剂治疗白细胞减少症的作用机制OA
Study on the mechanism of Shengxuebao Mixture in treating leukopenia based on network pharmacology and molecu-lar docking
目的:旨在运用网络药理学和分子对接技术,探讨生血宝合剂治疗白细胞减少症的作用机制.方法:生血宝合剂活性成分和靶点通过TCMSP数据库筛选,在GeneCards和Disgenet数据库中检索与白细胞减少症有关的靶点.通过交集分析得出共同靶点,并使用Cytoscape3.10.0软件构建"中药-活性成分-靶点"网络.为进一步进行拓扑分析筛选核心靶点,通过在STRING数据库中构建蛋白相互作用网络.采用AutoDock Vina 1.2.7软件对核心活性成分和靶点进行分子对接,利用DAVID平台进行GO功能及KEGG通路富集分析.结果:获得91个生血宝合剂活性成分,303个药物与疾病共同靶点,槲皮素、木犀草素、β-谷甾醇可能是主要活性成分,TP53、AKT1、TNF、IL6是核心靶点;KEGG共富集186个条目,主要富集在Toll样受体、NF-κB、PI3K/AKT、TNF、IL-17信号通路;分子对接显示,核心靶点与核心活性成分结合性较好.结论:本文初步揭示了生血宝合剂通过多成分-多靶点交互-多通路发挥抗白细胞减少症作用,为探究其作用机制提供了理论基础.
Objective:To use network pharmacology and molecular docking technology to explore the mechanism of action of Shengxuebao Mixture in the treatment of leukopenia.Methods:Active ingredients and corresponding targets of Shengxuebao Mixture were screened via the TCMSP database.Targets associated with leukopenia were retrieved from GeneCards and Dis-GeNET databases.Common targets were obtained through intersection analysis,and Cytoscape3.10.0 software was used to con-struct a traditional Chinese medicine-active ingredient-target network.For further topological analysis to screen core targets,a pro-tein interaction network was constructed in the STRING database.AutoDock Vina 1.2.7 software was used for molecular docking of core active ingredients and targets,and the DAVID platform was used for GO function and KEGG pathway enrichment analy-sis.Results:A total of 91 active components of Shengxuebao Mixture were obtained.303 potential targets of Shengxuebao Mix-ture for the treatment of diseases were obtained.Quercetin,kaempferol,β-sitosterol may be the main active components for the ef-fects.TP53,AKT1,TNF and IL6 were identified as the core targets.KEGG enriched 186 items,and the main pathways were Toll-like receptors,NF-κB,PI3K/AKT,TNF,and IL-17 signaling pathway.The core active components were capable of effectively binding to the predicted core targets,as revealed by molecular docking analysis.Conclusion:These findings preliminarily eluci-date the multi-component,multi-target,and multi-pathway interactive effects of Shengxuebao Mixture against leukopenia,provid-ing a theoretical basis for further investigation into its mechanisms.
