目的:探究颅复丸对KM小鼠的急性毒性.方法:48只KM小鼠分为3组,分别为空白对照组和颅复丸高、低剂量组,分别予生理盐水和13.2、6.6 g·kg-1颅复丸灌胃,之后连续观察14 d,期间记录小鼠的一般状态、体质量及行为活动等,观察结束后取血清检测常规生化指标,解剖小鼠观察肝、心、脾、肺、肾形态并测定其脏器系数,并观察组织病理形态.结果:各组小鼠均无死亡,未见无明显中毒症状,各组小鼠体质量、脏器系数、脏器病理形态等方面无明显差异,生化检测中丙氨酸氨基转移酶(ALT)、白蛋白(ALB)、总蛋白(TP)、甘油三酯(TG)、尿素(UREA)等指标有不同程度升高,但差异无明显剂量依赖性.结论:13.2 g·kg-1的剂量范围以内,颅复丸灌胃对KM小鼠主要脏器未造成明显病理损伤,但用药过程中需监测其对肝肾功能的影响.该研究可为临床用药提供参考.

Objective:To investigate the acute toxicity of Lufu Pills(LFP)in Kunming(KM)mice.Methods:Forty-eight KM mice were randomly divided into three groups:a blank control group and high-and low-dose LFP groups.The control group received normal saline,whereas the treatment groups received LFP at 13.2 and 6.6 g·kg-1,respectively,via gavage.All animals were observed for 14 consecutive days,during which general condition,body weight and behavioral changes were recorded.At the end of the observation period,serum samples were collected for routine biochemical analyses.The mice were then necropsied.Liver,heart,spleen,lung and kidney were examined macroscopically,weighed to calculate organ coefficients,and processed for histopathological evaluation.Results:No deaths or overt signs of toxicity occurred in any group.No significant differences were observed among groups in body weight,organ coefficients,or histopathological findings.Although biochemical parameters such as ALT(alanine aminotransferase),TP(total protein),ALB(albumin),TG(triglycerides),and UREA(urea nitrogen)showed vari-able elevations,these changes lacked a clear dose-dependent relationship.Conclusion:Within the dose range of 13.2 g·kg-1,intra-gastric administration of LFP did not cause significant pathological damage to the major organs of KM mice.However,it is neces-sary to monitor its impact on liver and kidney functions during the medication process.This study can provide a reference for clini-cal medication.
