目的:探究痛风定胶囊对高尿酸血症(HUA)大鼠的降尿酸效果及对肝肾的保护作用.方法:采用乙胺丁醇+腺嘌呤混悬液灌胃建立HUA大鼠模型,灌胃痛风定胶囊低、中、高(164.4、493.3、1 479.9 mg·kg-1)进行干预,持续7 d.实验结束后,计算肾脏系数,测定各组大鼠血清尿酸(UA)、尿素氮(BUN)、肌酐(CRE)、天冬氨酸氨基转移酶(AST)、谷氨酸氨基转移酶(ALT)含量,以及肝脏中的黄嘌呤氧化酶(XOD)活性,采用苏木精-伊红染色法观察肝肾组织的病理学特征.结果:模型大鼠血清UA、BUN、CRE、AST、ALT、肝脏XOD活性及肾脏系数显著升高,并表现出肾脏肾小管扩张、肾小球萎缩以及肝脏中炎症细胞浸润、肝细胞轻度脂变和水肿等;中、高剂量痛风定胶囊能显著降低大鼠肾脏系数血清UA、BUN、CRE水平(P<0.05,P<0.01或P<0.001),高剂量痛风定胶囊能显著降低大鼠AST水平(P<0.05),低、中、高剂量痛风定胶囊能显著降低大鼠ALT水平(P<0.05,P<0.01或P<0.001);各剂量痛风定胶囊可改善HUA大鼠肝脏和肾脏的病理变化.结论:痛风定胶囊对急性HUA大鼠具有明确的降尿酸作用,其机制与抑制黄嘌呤氧化酶活性密切相关,同时对高尿酸血症所致的肝肾功能损伤具有良好的保护作用,可为痛风定胶囊扩大临床适应症提供参考.

Objective:To explore the protective effect of Tongfengding capsule(TFDC)against hyperuricemia(HUA)and liver and kidney injuries in rats.Methods:The HUA rat model was established by intragastric administration of ethambutol combined with adenine.Meanwhile,TFDC(164.4,493.3,1 479.9 mg·kg-1)were given by intragastric administration for seven days.After the experiment,the kidney indexes were calculated,and the levels of uric acid(UA),creatinine(CRE),blood urea nitrogen(BUN),aspartate aminotransferase(AST)and alanine aminotransferase(ALT)in the serum of rats in each group were determined.Additionally,the activities of xanthine oxidase(XOD)in the liver tissue were measured.The pathological charac-teristics of liver and kidney tissues were observed using hematoxylin-eosin(HE)staining method.Results:In model rats,se-rum UA,BUN,CRE,AST,ALT levels,hepatic XOD activity and renal coefficient increased markedly,accompanied by renal tubular dilatation and glomerular atrophy in kidney,as well as inflammatory cell infiltration,mild fatty degeneration and edema of hepatocytes in liver.Medium-and high-dose TFDC significantly reduced the renal coefficient,serum UA and BUN and CRE levels(P<0.05,P<0.01 or P<0.001).The high-dose group markedly decreased serum AST(P<0.05).While All doses of TFDC groups effectively lowered serum ALT(P<0.05,P<0.01 or P<0.001),pathological lesions of liver and kidney in HUA rats.Con-clusion:TFDC can effectively reduce the serum UA level in HUA rats,which may be associated with the inhibition of XOD ac-tivities in liver.Meanwhile,the preparation provides favorable protective effects against hepatic and renal injury induced by HUA,which offers experimental evidence for expanding its clinical indications.
