目的:明确淫羊藿中淫羊藿次苷Ⅱ(icariside Ⅱ)的抗炎活性,并揭示其潜在作用机制,为淫羊藿抗炎药效物质基础及创新药物研发提供实验依据.方法:以脂多糖(LPS)诱导小鼠巨噬细胞(RAW 264.7)建立体外炎症模型,设置空白对照组、模型对照组、阳性对照组、淫羊藿次苷Ⅱ低、中、高剂量组,检测细胞上清液中一氧化氮(NO)释放量,采用实时荧光定量PCR(RT-qPCR)检测炎症因子IL-1β、IL-6、TNF-α的mRNA表达水平,蛋白质免疫印迹法(Western blot)检测诱导型一氧化氮合酶(iNOS)和环氧化酶2(COX-2)及NF-κB通路相关蛋白表达.结果:淫羊藿次苷Ⅱ各剂量可使LPS诱导小鼠巨噬细胞RAW 264.7 IL-1β、IL-6、TNF-α的mRNA表达水平显著下调,剂量依赖性抑制NO生成,并显著降低iNOS、COX-2的蛋白表达和pIKK、pIκB以及p-p65的表达.结论:淫羊藿次苷Ⅱ对LPS诱导的RAW 264.7细胞炎症反应具有显著抑制作用,其机制可能与调控NF-κB信号通路、抑制炎症因子释放及iNOS、COX-2蛋白表达有关.

Objective:To clarify the anti-inflammatory activity of icariside Ⅱ from EpimedⅡ Folium and reveal its underly-ing mechanism,so as to provide experimental evidence for the anti-inflammatory pharmacodynamic material basis of EpimedⅡFolium and the research and development of innovative drugs.Methods:An in vitro inflammatory model was established by in-ducing mouse RAW 264.7 macrophages with lipopolysaccharide(LPS).Blank control group,model control group,positive con-trol group,and low-,medium-and high-dose groups of icariside Ⅱ were set up.The release level of nitric oxide(NO)in cell super-natant was detected using a nitric oxide assay kit.The mRNA expression levels of inflammatory factors IL-1β,IL-6 and TNF-αwere detected by quantitative real-time PCR(RT-qPCR).The release of nitric oxide(NO)was measured by Griess reagent assay.The protein expression of inducible nitric oxide synthase(iNOS),cyclooxygenase-2(COX-2)and NF-κB pathway-related proteins was determined by Western blot.Results:All tested doses of icariside Ⅱ markedly downregulated the mRNA expression levels of IL-1β,IL-6 and TNF-α in LPS-stimulated RAW 264.7 murine macrophages,suppressed NO production in a dose-dependent man-ner,and significantly reduced the protein expressions of iNOS and COX-2,as well as the expression of p-IKK,p-IκB and p-p65.Conclusion:Icariside Ⅱ exerts a significant inhibitory effect on LPS-induced inflammatory responses in RAW 264.7 cells.Its mechanism may be related to the regulation of NF-κB signaling pathway,the inhibition of inflammatory factor release,and the downregulation of iNOS and COX-2 protein expression.
