花椒养生汤提取工艺优化研究OA
Optimization of the extraction of Huajiao Yangsheng decoction
目的:系统分析花椒养生汤(YST)的化学成分组成,优化YST提取工艺.方法:采用UPLC-Q-Orbitrap HRMS分析YST化学成分;以人参皂苷Rg1、Rb1、Re,肉桂醛,姜酚和羟基-α-山椒素为指标建立质量标准,以YST中上述成分含量综合得分为评价指标,考察乙醇用量、提取时间、提取次数和浸泡时间等因素,基于正交设计对YST的提取工艺参数进行优化.结果:从YST中鉴别48种化合物,包括人参皂苷Re和6-姜酚等;正交设计结果确定YST最佳提取工艺为浸泡2 h后加入8倍量70%乙醇回流提取3次,每次1.5 h.结论:本研究明确了YST化学成分组成,建立了YST质量标准并优化其提取工艺,为其临床应用和现代剂型开发奠定了坚实基础.
Objective:To systematically analyze the chemical constituents of Huajiao Yangsheng decoction(YST)and opti-mize its extraction of the YST.Methods:UPLC-Q-Orbitrap HRMS was used to identify the chemical constituents of YST.HPLC was used to establish quality control method using the reference agents of ginsenosides Rg1,Rb1,Re,cinnamaldehyde,gingerol,and hydroxy-α-sanshool(HAS).The weighted comprehensive score regarding the above-mentioned reference agents in YST was used as index,and then orthogonal design was used to optimize the YST extraction with the ethanol content,soaking time,extrac-tion time,extraction frequency.Results:A total of 48 compounds were identified from YST,including ginsenoside Re,6-gingerol,etc.The optimal extraction parameters for YST based on orthogonal design was decided as:soaking for 2 h,8 times 70%alcohol for reflux extraction 3 times,and each time lasting for 1.5 h.Conclusion:This study identifies chemical composition profile of YST,establishes its quality standards,and optimizes its extraction,laying the foundation for its clinical application and modern dosage form development.
