基于多能量-多模式高分辨质谱的蔗糖酯结构鉴定策略及其在锦灯笼中的应用OA
Strategy for structural identification of sucrose esters based on multi-energy and multi-mode HRMS and its applica-tion in Physalis alkekengi L.var.franchetii(Mast.)Makino
目的:本研究旨在建立基于高分辨质谱的蔗糖酯结构鉴定策略,实现对该类成分的快速、准确解析.方法:采用UPLC-Q-Orbitrap-HRMS技术,结合不同碰撞能量与正负离子模式,对11个已知蔗糖酯单体化合物进行系统分析,考察碰撞能量、离子模式与碎片离子之间的对应关系,归纳其质谱裂解规律,并将其进一步应用于锦灯笼提取物中未知蔗糖酯成分的结构推测.结果:基于不同位置的酯键在不同碰撞能量下的差异化断裂倾向与顺序,构建了针对性的结构鉴定策略.从锦灯笼中共推断出35个未知蔗糖酯,包括23个双/三取代蔗糖酯与12个2,3,4,6-四取代蔗糖酯,其中四取代蔗糖酯中各取代基的连接位置均得以明确指认,双/三取代蔗糖酯仅确认取代基组合,具体连接位置尚未明确.结论:所建立的多能量、多模式高分辨质谱分析策略,可为2,3,4,6-四取代蔗糖酯的结构鉴定提供系统、可靠的方法依据,有效解决该类成分因同分异构体多、结构相似导致的鉴定难题,为蔗糖酯类成分的结构表征与深入研究提供普适性的技术参考.
Objective:This study aimed to establish a structural identification strategy for sucrose esters based on high-resolution mass spectrometry(HRMS)to achieve rapid and accurate analysis of these compounds.Methods:UPLC-Q-Orbitrap-HRMS technology was employed,combined with different collision energies and positive/negative ion modes,to systematically analyze 11 known sucrose ester monomers.The corresponding relationships between collision energy,ion mode and fragment ions were investigated.Their mass spectral fragmentation rules were summarized,and this strategy was further applied to the structural speculation of unknown sucrose ester components in Physalis alkekengi L.var.franchetii(Mast.)Makino.Results:Based on the differential cleavage tendency and sequence of ester bonds at different positions under different collision energies,a targeted struc-tural identification strategy was constructed.A total of 35 previously unknown sucrose esters were inferred from the extract,in-cluding 23 di-/tri-substituted and 12 2,3,4,6-tetra-substituted sucrose esters.The specific substitution positions of all tetra-substituted sucrose esters were unambiguously assigned,while for the di-/tri-substituted ones,only the substituent composition could be confirmed,with their specific locations remaining undetermined.Conclusion:The established multi-energy and multi-mode high-resolution mass spectrometry analysis strategy can provide a systematic and reliable method basis for the structural identification of 2,3,4,6-tetra-substituted sucrose esters,effectively solving the identification difficulties of such components caused by numerous isomers and similar structures,and pro-viding a universal technical reference for the structural charac-terization and in-depth research of sucrose ester components.
