不同产区丹参的农艺性状与有效成分综合评价OA

Comprehensive Evaluation of Agronomic Traits and Active Components of Salvia miltiorrhiza from Different Producing Areas

目的:比较不同产区丹参Salvia miltiorrhiza Bge.种质的根部农艺性状与主要有效成分差异,开展综合评价并筛选优异种质.方法:收集不同产区 50 份丹参种质,同时纳入 3 份近缘野生对照材料(云南鼠尾草S.yunnanensis C.H.Wright、华鼠尾草S.chinensis Benth.),将其进行统一栽培与田间管理;测定根部关键农艺性状,包括根鲜质量、根长、粗根直径、皮木比、根数、三级根数;采用UPLC测定丹参素钠、丹酚酸B、隐丹参酮、丹参酮Ⅰ、丹参酮ⅡA质量分数,并计算总丹参酮(隐丹参酮、丹参酮Ⅰ、丹参酮ⅡA)质量分数.运用描述统计、相关性分析、聚类分析、主成分分析(PCA)、正交偏最小二乘法-判别分析(OPLS-DA)结合变量重要性投影(VIP)筛选特征指标;以根鲜质量、三级根数、皮木比,以及丹参素钠、丹酚酸B和总丹参酮质量分数 6 个指标构建综合评价体系,结合熵权-逼近理想解排序(TOPSIS)法完成综合评价.结果:不同产区丹参种质性状差异明显,根鲜质量为 71.85~1 146.54 g,丹酚酸B质量分数为 3.535%~7.844%,总丹参酮质量分数为 0.038%~0.933%;3 份近缘野生对照材料与栽培丹参在根系结构、有效成分积累方面存在明显差异;相关性分析表明,三级根数、皮木比等根系结构性状与总丹参酮积累存在相关性;PCA、OPLS-DA和VIP分析结果显示,有效成分与根系性状共同决定种质间差异;熵权-TOPSIS评价结果显示,山东平邑(SD-PYI-01)、山东鄄城(SD-JCH-01)、山东沂源(SD-YYU-01)种质的综合表现最优.结论:不同产区丹参种质的农艺性状与主要有效成分含量差异明显;根系结构性状可作为丹参品质初筛的辅助依据;基于 6 个指标构建的综合评价体系适用于丹参优异种质筛选.

Objective:To compare differences in root agronomic traits and major active components among Salvia miltiorrhiza germplasm resources from different producing areas,to conduct a comprehensive evaluation,and to screen superior germplasms.Methods:Fifty S.miltiorrhiza germplasm resources were collected from different origins,together with three closely related wild control samples,including S.yunnanensis and S.chinensis.These samples were subjected to uniform cultivation and field management.Then,the following parameters were measured:key root agronomic traits(fresh root weight,root length,thick-root diameter,phloem-to-xylem ratio,root number,and tertiary root number),as well as the mass fractions of sodium danshensu,salvianolic acid B,cryptotanshinone,tanshinone Ⅰ,and tanshinone ⅡA by ultra-performance liquid chromatography(UPLC),with total tanshinone(cryptotanshinone,tanshinone Ⅰ,and tanshinone ⅡA)mass fractions calculated.Characteristic indicators were screened by descriptive statistics,correlation analysis,cluster analysis,principal component analysis(PCA),orthogonal partial least squares-discriminant analysis(OPLS-DA),and variable importance in projection(VIP).Subsequently,a comprehensive evaluation system was constructed using six indicators,namely fresh root weight,tertiary root number,phloem-to-xylem ratio,along with the mass fractions of sodium danshensu,salvianolic acid B,and total tanshinone.Finally,a comprehensive evaluation was performed by combining the entropy-weighted technique for order preference by similarity to an ideal solution(TOPSIS)method.Results:Significant differences were observed among S.miltiorrhiza germplasm resources from different producing areas,with fresh root weight ranging from 71.85 g to 1 146.54 g,salvianolic acid B mass fraction ranging from 3.535%to 7.844%,and total tanshinone mass fraction ranging from 0.038%to 0.933%.The three closely related wild control samples differed significantly from cultivated S.miltiorrhiza in root structure and active component accumulation.Correlation analysis revealed an association between total tanshinone accumulation and root structural traits such as tertiary root number and phloem-to-xylem ratio.PCA,OPLS-DA,and VIP analyses indicated that differences among germplasm resources were co-determined by active components and root traits.Entropy-weighted TOPSIS evaluation showed that the germplasm resources with top comprehensive performance were from Pingyi,Shandong(SD-PYI-01),Juancheng,Shandong(SD-JCH-01),and Yiyuan,Shandong(SD-YYU-01).Conclusion:S.miltiorrhiza germplasm resources from different producing areas demonstrate significant differences in agronomic traits and major active component contents.Root structural traits can serve as auxiliary criteria for preliminary quality screening.Furthermore,the comprehensive evaluation system constructed based on six indicators is applicable for screening superior germplasm.
