目的:明确不同产区甘草药材基原与v种群遗传多样性,解析影响甘草有效成分积累的关键因素,为甘草药材规范化种植与质量管控提供依据.方法:简单重复区间序列(ISSR)利用分子标记技术分析样品遗传特征;采用DNA条形码技术开展基原鉴定;采用HPLC测定甘草中 8 个有效成分的含量;通过药用植物全球产地生态适宜性区划信息系统获取 19 个气候因子数据,采用曼特尔相关性分析、灰色关联度分析揭示遗传因子、气候因子对不同基原甘草有效成分积累的影响.结果:经鉴定,受试样品基原为甘草或光果甘草;甘肃、内蒙古所产甘草居群遗传多样性较丰富;气温、降水等气候因子和等位基因数、基因多样性指数等遗传参数与甘草药材中有效成分含量呈正相关;气候因子是甘草有效成分积累的主导因子,遗传因子是光果甘草中有效成分积累的主导因子.结论:本研究明确了不同产区甘草药材的基原,以及遗传因子、气候因子对 2 种基原甘草有效成分含量的影响,为其质量评价和道地性研究提供了方法参考.

Objective:To clarify the botanical origin and population genetic diversity of Glycyrrhizae Radix et Rhizoma from different production areas and analyze the key factors affecting the accumulation of active components,thereby providing a basis for standardized cultivation and quality control of Glycyrrhizae Radix et Rhizoma.Methods:Inter-simple sequence repeat(ISSR)molecular markers were employed to analyze the genetic characteristics of the samples.DNA barcoding was used for the identification of botanical origin.The content of eight active components in Glycyrrhizae Radix et Rhizoma was determined by high performance liquid chromatography(HPLC).The data of 19 climatic factors were obtained from the Geographic Information System for Global Medicinal Plants(GMPGIS).Mantel correlation analysis and grey relational analysis were performed to reveal the effects of genetic factors and climatic factors on the accumulation of active components in Glycyrrhizae Radix et Rhizoma from different botanical origins.Results:The tested samples belonged to two species:Glycyrrhiza uralensis and G.glabra.The populations from Gansu and Inner Mongolia exhibited rich genetic diversity.Climatic factors such as temperature and precipitation,as well as genetic parameters including the number of alleles and gene diversity index,showed positive correlations with the content of active components in Glycyrrhizae Radix et Rhizoma.Climatic factors were the dominant factors affecting the accumulation of active components in G.uralensis,whereas genetic factors mainly influenced the accumulation of active components in G.glabra.Conclusion:This study clarifies the botanical origin of Glycyrrhizae Radix et Rhizoma from different production areas and the effects of genetic and climatic factors on the active component content of the two species,providing a methodological reference for quality evaluation and geoherbalism research of Glycyrrhizae Radix et Rhizoma.
