基于网络药理学和分子对接技术探讨甘草附子汤治疗膝骨关节炎作用机制OA

Exploring the Mechanism of Gancao Fuzi Decoction in Treating Knee Osteoarthritis Based on Network Pharmacology and Molecular Docking Technology

目的 已有证据表明甘草附子汤治疗膝骨关节炎具有良好的临床疗效.为进一步探究其潜在作用机制及相关信号通路,采用网络药理学与分子对接技术进行分析.方法 通过中药系统药理学数据库与分析平台(https://old.tcmsp-e.com/tcmsp.php)检索甘草附子汤的活性成分及其对应靶点;从GeneCards数据库(https://www.genecards.org/)和OMIM数据库(https://www.omim.org/)中收集膝骨关节炎相关基因.利用STRING数据库(https://string-db.org/)和Cytoscape软件构建蛋白质-蛋白质相互作用(protein-protein interaction,PPI)网络,筛选核心靶点,并绘制"中药-成分-靶点-疾病"网络图.基于R语言Bioconductor平台中的相关数据包进行基因本体(gene ontology,GO)功能注释与京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析.最后,运用分子对接技术评估主要活性成分与核心靶标白细胞介素-6(interleukin-6,IL-6)、RAC-α丝氨酸/苏氨酸蛋白激酶(RAC-α serine/threonine-protein kinase,Akt1)及肿瘤坏死因子(tumor necrosis factor,TNF)之间的结合亲和力.结果 从中药系统药理学数据库与分析平台中共筛选出1028种化合物,对应191个甘草附子汤非重复靶点,获得与抗膝骨关节炎相关的潜在交叉靶点115个.PPI网络分析显示,核心靶点包括IL-6、TNF、Akt1、白细胞介素-1β、基质金属蛋白酶-9、前列腺素内过氧化物合酶-2、胱天蛋白酶-3、转录因子AP-1、缺氧诱导因子-1α和表皮生长因子受体.GO功能注释与KEGG通路富集分析提示,这些靶点主要参与对脂多糖的反应、对细菌来源分子的反应以及活性氧代谢过程等生物学过程;相关信号通路主要包括白细胞介素-17信号通路、TNF信号通路和缺氧诱导因子-1信号通路.分子对接结果表明,主要活性成分与IL-6、TNF、Akt1均具有良好的结合亲和力,其中与TNF的结合最为显著,提示TNF可能是甘草附子汤抗膝骨关节炎作用中的关键靶点.结论 甘草附子汤可能通过调节脂多糖反应、细菌来源分子反应及活性氧代谢过程等途径缓解膝骨关节炎,揭示了该方治疗膝骨关节炎的潜在分子机制,为其进一步研究及新药开发提供了依据.

Objective Evidence suggests that Gancao Fuzi Decoction(GCFZD)demonstrates good clinical efficacy in treating knee osteoarthritis(KOA).To further explore its underlying mechanisms and signaling pathways,this study employed network pharmacology and molecular docking technology for investigation.Methods Active components of GCFZD and their corresponding target proteins were retrieved from the TCMSP database.KOA-related genes were collected from the GeneCards and OMIM databases.The STRING database and Cytoscape software were used to construct a protein-protein interaction(PPI)network,identify core targets,and build a"Herb-Component-Target-Disease"network diagram.Functional enrichment analysis,including gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)pathway analyses,were carried out via relevant software packages based on the Bioconductor platform of R language.Finally,molecular docking was utilized to evaluate the binding affinity between the main active components and the core targets interleukin-6(IL-6),RAC-alpha serine/threonine-protein kinase(Akt1),and tumor necrosis factor(TNF).Results A total of 1028 compounds were screened from the TCMSP database,corresponding to 191 unique potential targets of GCFZD.Cross-analysis identified potential overlapping targets for GCFZD against KOA.PPI network analysis revealed that the core targets associated with KOA treatment included IL-6,TNF,Akt1,interleukin-1 beta(IL-1β),matrix metalloproteinase-9(MMP-9),prostaglandin-endoperoxide synthase 2(PTGS2),caspase-3(CASP3),transcription factor AP-1(JUN),hypoxia-inducible factor 1-alpha(HIF-1A),and epidermal growth factor receptor(EGFR).GO and KEGG enrichment analyses indicated that the primary biological processes involved were the response to lipopolysaccharide,response to molecule of bacterial origin,and the reactive oxygen species metabolic process.The most significantly enriched signaling pathways were the IL-17 signaling pathway,TNF signaling pathway,and HIF-1 signaling pathway.Molecular docking results showed that the main active components had good binding affinity to IL-6,TNF,and Akt1,with the binding to TNF being particularly strong,suggesting that TNF might be the most crucial target for GCFZD's anti-KOA effects.Conclusion GCFZD may alleviate KOA by modulating the response to lipopolysaccharide,the response to molecules of bacterial origin,and the reactive oxygen species metabolic process.Our study reveals the potential molecular mechanism of GCFZD in treating KOA,demonstrating its value for further research and its potential as a new drug candidate.
